Cytokines, chemokines, and growth factors are key mediators of cell proliferation, migration, and immune response, and in tumor microenvironment, such factors contribute to regulation of tumor growth, immune cell recruitment, angiogenesis, and metastasis. In body fluids, levels of inflammatory mediators reflect the patient immune response to the disease and may predict the effects of targeted therapies. Significant improvements in cytokine detection techniques have been made during last 10 years leading to sensitive quantification of such potent molecules present in low pg/mL levels. Among the techniques, Luminex xMAP multiplex assays allow for simultaneous quantification of up to 100 analytes with high sensitivity, broad dynamic range of quantification, high throughput, and minimal sample requirements. In this chapter we describe a detailed protocol for the application of xMAP assays using Luminex 200™ analyzer with xPonent acquisition software to quantify cytokines, chemokines, and growth factors secreted to blood serum and plasma of cancer patients. We also discuss how sample preparation, instrument settings, and standard curve fitting algorithms can influence validity of obtained results. Special attention is paid to data analysis using open source R statistical environment and we provide an example dataset of cytokine levels measured in serum and corresponding R script for standard curve fitting and concentration estimates.

Background: The mechanisms underlying the beneficial effects of exercise on the human placenta are poorly understood. The objective of the current study is to ascertain the influence of a supervised concurrent exercise intervention from gestational week 17 until birth on key cytokines involved in placental development and function. Secondary aims were to explore: (a) the moderating effects of fetal sex and maternal weight status; and (b) whether gestational weight gain, lifestyle behaviors (diet, sleep patterns, and physical activity), and physical fitness (strength and cardiorespiratory fitness) mediated the effects of exercise on placental cytokines.

Background: Alzheimer's disease (AD) is characterized by a complex interplay between amyloid-β (Aβ) and tau pathologies, with increasing evidence implicating cerebral blood flow (CBF) alterations as a critical, yet underexplored, contributor to disease progression. This study aimed to investigate the associations between regional CBF and cerebrospinal fluid (CSF) biomarkers- Aβ1-42, total tau (T-Tau), and phosphorylated tau (P-Tau181)-across the AD continuum.
Methods: We conducted a cross-sectional analysis using data from 416 participants enrolled in the Alzheimer's Disease Neuroimaging Initiative (ADNI), including cognitively normal individuals, patients with mild cognitive impairment (MCI), and those with AD.
SARS-CoV-2 population-based seroprevalence surveys are useful for estimating the extent of SARS-CoV-2 infections, which may be underestimated by COVID-19 case counts. In endometrial cancer, detection of oncogenic mutations in the polymerase epsilon (POLE) gene is crucial for accurate staging according to the 2023 International Federation of Gynecology and Obstetrics classification and for minimizing overtreatment. However, POLE sequencing is expensive, time-consuming, and often inaccessible in settings without specialized equipment. We developed a novel multiplex kit for the detection of POLE mutations using a Luminex (xMAP) assay in a single reaction.
