Leucine aminopeptidase (LAP) is an essential proteolytic enzyme and potential biomarker for liver malignancy. Overexpression of LAP is directly linked with some fatal physiological and pathological disorders. In this regard, we have designed an activity based electrochemical substrate leucine-benzyl ferrocene carbamate (Leu-FC) for selective profiling of LAP activity in live cells. In practice, LAP instantaneously hydrolyze the Leu residue of the substrate Leu-FC to eliminate the unmasked electrochemical reporter amino ferrocene via predefined self-immolative cascade. The electrochemical signal is distinctly specific for LAP and free of other electroactive biological interference. The substrate Leu-FC empowered sensor displayed broad dynamic range with admirable detection limits. On top of this, the probe Leu-FC was employed in real-time active profiling of cellular LAP activity in HepG2 cells and effect of LAP inhibitor. In extent, the substrate Leu-FC can effectively monitor cisplatin induced overexpression of LAP activity in HepG2 cells in presence and absence of bestatin. The sensor showcased an excellent reliability towards monitoring cellular LAP activity in HepG2 cells. Unlike the traditional antibody-based immunoassays, our approach is capable of monitoring in-situ activity of LAP in live cells.

Objective: Therapeutic potential of selective aggrecanase inhibition in osteoarthritis (OA) was previously demonstrated using a variant of endogenous tissue inhibitor of metalloproteinase-3 (TIMP-3); however, this relied on transgenic mice overexpressing TIMP-3. Here, we develop a translational approach for harnessing the aggrecanase-selective inhibitory activity of TIMP-3 using the latency associated peptide (LAP) technology.
Methods: We successfully produced and purified recombinant LAP-TIMP-3 fusion proteins and determined the pharmacokinetics of these proteins in vivo following systemic injection.

Background/objective: Cyclosporine A and other calcineurin inhibitors have been identified as prospective treatments for preventing Alzheimer's disease. We previously found that calcineurin inhibitors elicit a unique behavioral profile in zebrafish larvae, characterized by increased activity, acoustic hyperexcitability, and reduced visually guided behaviors. Screening a large library of FDA-approved compounds using Z-LaP Tracker revealed that some heart medications produce a similar behavioral profile, suggesting these drugs may exert calcineurin-inhibitor-like effects relevant to prevent-ing or ameliorating Alzheimer's disease.

Background: Pacing in ultra-triathlon has been investigated by analyzing lap times from Double to Deca Iron ultra-triathlon for World Cup races but not for a World Championship. The present study aimed to investigate pacing in ultra-triathletes competing in the fastest and largest World Championship in Quintuple Iron ultra-triathlon ever held in history.
Methods: A total of 11 female and 24 male finishers who completed the 2024 Quintuple Ultra Triathlon World Championship in Colmar, France, were analyzed.

Aluminum (Al) toxicity has attracted widespread attention owing to its bioavailability, environmental persistence, and detrimental impacts on human health. It is primarily absorbed into the systemic circulation via the gastrointestinal tract, which may damage the brush border membrane (BBM) and intestinal mucosal barrier, leading to numerous adverse health effects. Therefore, the present in vivo and in vitro studies were carried out to evaluate the effect of aluminum chloride (AlCl) treatment on oxidative stress parameters, cellular metabolism, membrane integrity, and DNA damage in the rat intestine.
