Background: This study was a multicenter, parallel-group, double-blind, double-dummy, randomized, noninferiority trial to evaluate the efficacy and safety of γ-linolenic acid (GLA) relative to α-lipoic acid (ALA) over a 12-week treatment period in type 2 diabetes mellitus (T2DM) patients with painful diabetic peripheral neuropathy (DPN).
Methods: This study included 100 T2DM patients between 20 and 75 years of age who had painful DPN and received either GLA (320 mg/day) and placebo or ALA (600 mg/day) and placebo for 12 weeks. The primary outcome measures were mean changes in pain intensities as measured by the visual analogue scale (VAS) and the total symptom scores (TSS).
Results: Of the 100 subjects who initially participated in the study, 73 completed the 12-week treatment period. Per-protocol analyses revealed significant decreases in the mean VAS and TSS scores compared to baseline in both groups, but there were no significant differences between the groups. The treatment difference for the VAS (95% confidence interval [CI]) between the two groups was -0.65 (-1.526 to 0.213) and the upper bound of the 95% CI did not exceed the predefined noninferiority margin (δ₁=0.51). For the TSS, the treatment difference was -0.05 (-1.211 to 1.101) but the upper bound of the 95% CI crossed the noninferiority margin (δ₂=0.054). There were no serious adverse events associated with the treatments.
Conclusion: GLA treatment in patients with painful DPN was noninferior to ALA in terms of reducing pain intensity measured by the VAS over 12 weeks.

This study investigates a multifunctional hydrogel system integrating carboxymethyl cellulose (CMC) in a 3D-printed limonene (LIM) scaffold coated with poly(3,4-ethylenedioxythiophene): polystyrene sulfonate (PEDOT:PSS). The system allows to enhance wound healing, prevent infections, and monitor the healing progress. CMC is crosslinked with citric acid (CA) to form the hydrogel matrix (CMC-CA), while the 3D-printed limonene (LIM) scaffold is embedded within the hydrogel to provide mechanical support.

A fused octapyrrolylanthracene, representing a nonplanar pyrrole-fused aza-nanographene with two deep gulf-edge regions, was readily synthesized and found to exhibit a ladder-shaped bent structure. Electrochemical studies revealed reversible multielectron oxidation up to four electrons. Stepwise oxidation with AgPF or I afforded a singlet diradical dication and a closed-shell aromatic tetracation.

It is helpful for diagnostic purposes to improve our current knowledge of gut development and serum biochemistry in young piglets. This study investigated serum biochemistry, and gut site-specific patterns of short-chain fatty acids (SCFA) and expression of genes related to barrier function, innate immune response, antioxidative status and sensing of fatty and bile acids in suckling and newly weaned piglets. The experiment consisted of two replicate batches with 10 litters each.

Importance: Pregnant individuals with polycystic ovary syndrome (PCOS) present with a higher risk of pregnancy complications, including gestational diabetes, preeclampsia, and preterm birth. Myo-inositol supplementation may reduce these risks.
Objective: To determine whether daily supplementation with myo-inositol during pregnancy among individuals with PCOS reduces the risk of a composite outcome of gestational diabetes, preeclampsia, and preterm birth.

High-fat foods are decomposed into fatty acids during digestion and absorption, primarily occurring in the gastrointestinal tract, and numerous studies have indicated that long-term high-fat diets significantly increase the incidence of intestinal disorders. As a critical intestinal hormone, serotonin (5-hydroxytryptamine, 5-HT) is involved in regulating intestinal peristalsis, secretion, and visceral sensitivity. However, due to the lack of methods capable of reproducing intestinal mechanical activities and in situ monitoring of 5-HT levels, the influence of high-fat diets on intestinal 5-HT release remains unclear.
