Vitamin D has been proposed to attenuate chemotherapy-induced gastrointestinal mucositis (GM). In the intestine, local catabolism of active vitamin D [1,25-dihydroxyvitamin D₃] is mediated by the enzyme Cyp24a1. This study assessed whether deletion of Cyp24a1 specifically in intestinal epithelial cells can protect against 5-fluorouracil (5-FU)-induced intestinal injury and microbiome disruption in mice.

Small extracellular vesicles (sEVs) critically orchestrate inter-tissue and inter-organ communications and may play essential roles in heart-tumor interaction. However, whether cancer-secreted sEVs affect the progression of doxorubicin-induced cardiotoxicity (DOXIC) via orchestrating the tumor cell-cardiomyocyte crosstalk has not yet been explored. Herein, we reveal that Doxorubicin (DOX)-treated breast cancer cells secrete sEVs (D-BCC-sEVs) that exacerbate DOX-induced ferroptosis of human iPSC-derived cardiomyocytes (hiCMs).

Radioresistance poses a significant obstacle in the management of Non-Small Cell Lung Cancer (NSCLC), often diminishing the effectiveness of radiotherapy and leading to treatment failures and adverse clinical outcomes. This study develops radioresistant NSCLC models, revealing that Secreted Protein Acidic and Rich in Cysteine (SPARC) as a crucial modulator of this resistance, through the inhibition of ferroptosis. To address this radioresistance, we propose a novel ferroptosis-oriented radiosensitization strategy specifically designed to enhance radiotherapy effectiveness in radioresistant NSCLC.
