Article Abstract

Herein, we describe a protocol for the preparation and analysis of primary isolated rat hepatocytes in a 3D cell culture format described as spheroids. The hepatocyte cells spontaneously self-aggregate into spheroids without the need for synthetic extracellular matrices or hydrogels. Primary rat hepatocytes (PRHs) are a readily available source of primary differentiated liver cells and therefore conserve many of the required liver-specific functional markers, and elicit the natural in vivo phenotype when compared with common hepatic cells lines. We describe the liquid-overlay technique which provides an ultra-low attachment surface on which PRHs can be cultured as spheroids. © 2019 The Authors. The development of biomimetic scaffolds that emulate the extracellular matrix (ECM) is critical for advancing cell-based therapies and tissue regeneration. This study reports the formulation of CHyCoGel, a novel injectable, ECM-mimetic hydrogel scaffold composed of chitosan, hyaluronic acid, chondroitin sulfate, and an amphiphilic stabilizer. CHyCoGel addresses key limitations of existing scaffolds, offering improved structural uniformity, injectability, and gelation suitable for cell encapsulation and minimally invasive delivery.

Food combinations featuring specific functional components represent one of the effective intervention strategies for alleviating functional gastrointestinal disorders induced by dietary and environmental factors. Honey and aloe vera have both been recognized as natural agents with laxative effects, yet the synergistic effects of their combination in alleviating constipation and the underlying regulatory mechanism remain to be elucidated. This study formulated a honey-aloe paste by employing honey as the primary ingredient compounded with aloe vera gel and investigated its preventive effects on loperamide-induced slow-transit constipation through a comprehensive analysis of gastrointestinal function and intestinal microenvironment.

We previously demonstrated the CFTR correctors VX-445 (elexacaftor) and S-VX-121 (vanzacaftor) potentiate heterologously-expressed BK channels, as well as in primary human bronchial epithelial cells (HBEs). This potentiation of BK resulted in altered vasoreactivity and neuronal excitability. We postulated novel compounds could be identified that would potentiate BK while not affecting CFTR.
