Article Abstract

Lessons Learned: Pharmacokinetics characteristics of niraparib in Chinese patients were similar to those in white patients. Niraparib could be well tolerated by Chinese patients, and adverse events were manageable in this study. Population pharmacokinetics analysis indicated that baseline body weight had a modest impact on pharmacokinetics parameters of niraparib; however, it was not considered clinically important.

Background: This randomized, open-label, single-arm, phase I study was designed to investigate the pharmacokinetics (PK) and safety of niraparib in Chinese patients with epithelial ovarian cancer.

Methods: Eligible patients were randomized in a 1:1:1 ratio to receive 100, 200, or 300 mg of niraparib once daily. PK parameters were analyzed after single and multiple dose administrations.

Results: Thirty-six Chinese patients were enrolled in total. Niraparib was rapidly absorbed after administration, and median time-to-peak (T ) was 3 hours. The long terminal elimination half-life (T ∼ 35 hours) supports once-daily dosing regimen. The exposure to niraparib showed linear and dose-proportional pharmacokinetics, whereas other PK parameters such as T , T , and accumulation ratio were dose independent. Population PK analysis indicated that there was no effect of race on niraparib PK parameters, whereas baseline body weight had a modest impact on niraparib exposure. Grade 3/4 treatment-emergent adverse events (TEAEs; reported in ≥10% of patients) included platelet count decreased (a total of five patients who were all from the 300-mg group) and neutrophil count decreased. The TEAEs were manageable after dose modification.

Conclusion: The PK profile of niraparib in Chinese patients is consistent with that in white patients. Niraparib is safe and well tolerated in Chinese patients with ovarian cancer.

Background: Moderate to severe acne scars pose a therapeutic challenge and often require multimodal treatments. Ablative fractional laser (AFL) and fractional microneedling radiofrequency (FMR) are commonly used, but their alternating application has not been fully studied.

Patients And Methods: In this prospective, randomized, single-blind, split-face trial, 20 patients (Fitzpatrick III-IV) received four treatments at 4-week intervals: one facial side underwent alternating FMR (sessions 1, 3) and AFL (sessions 2, 4), while the contralateral side received AFL alone.

This study aimed to evaluate the quality of multidisciplinary team (MDT) management in healthcare-associated infection (HAI) prevention and control, as well as its impact on multidrug-resistant organism (MDRO) infections. This was a retrospective, single-center study with a small sample size. A total of 400 patients admitted to the Departments of Critical Care Medicine or Orthopedics between January 2022 and December 2023 were divided into a control group (n = 200, receiving conventional HAI management) and an experimental group (n = 200, undergoing MDT management).

Objective: To expand the clinical phenotype associated with MYRF mutations in disorders of sex development (DSDs).

Methods: We present a case of a 17-year-old patient with a female phenotype who presented with primary amenorrhea.

Results: The patient's external genitalia was entirely female in appearance, though there was no opening of vagina below the orifice of urethra.

Asthma is a chronic respiratory disorder characterized by airway inflammation, hyperresponsiveness, and reversible airflow obstruction. Despite therapeutic strategies, asthma remains inadequately controlled in many patients. Genetic predisposition plays a significant role in asthma pathogenesis, and the Proteinase-Activated Receptor 2 (PAR-2), encoded by the F2RL1 gene, has been associated with asthma.
