Stem cells balance cellular fates through asymmetric and symmetric divisions in order to self-renew or to generate downstream progenitors. Symmetric commitment divisions in stem cells are required for rapid regeneration during tissue damage and stress. The control of symmetric commitment remains poorly defined. Using single-cell RNA sequencing (scRNA-seq) in combination with transcriptomic profiling of HSPCs (hematopoietic stem and progenitor cells) from control and mA methyltransferase Mettl3 conditional knockout mice, we found that mA-deficient hematopoietic stem cells (HSCs) fail to symmetrically differentiate. Dividing HSCs are expanded and are blocked in an intermediate state that molecularly and functionally resembles multipotent progenitors. Mechanistically, RNA methylation controls Myc mRNA abundance in differentiating HSCs. We identified MYC as a marker for HSC asymmetric and symmetric commitment. Overall, our results indicate that RNA methylation controls symmetric commitment and cell identity of HSCs and may provide a general mechanism for how stem cells regulate differentiation fate choice.

Mutual grooming, where both participants groom each other simultaneously, is a type of allogrooming and probably also plays an important role as a social and hygienic function; however, it has attracted little attention, probably due to its rarity compared to prevailing unidirectional grooming in many social animals, such as primate species. While previous studies have shown that mutual grooming has functional significance, such as promoting social bonds and maximizing short-term benefits, we do not know why mutual grooming is much rarer than unidirectional grooming, and its restrictive factors have been mostly uninvestigated. In this article, we propose a hypothesis of cognitive constraints in which cognitive complexity is assumed as an underpinning mechanism for mutual grooming, thereby restricting its prevalence across animal species.

In view of the limited mineral resources, it is imperative to develop coordinated strategies to mitigate the risks of information asymmetry in the e-waste recyclable supply chain. This study explores the two-period strategies employed by brand owners when outsourcing to qualified recyclers who engage in both processing and remanufacturing, and operate across online and offline waste recycling channels, under conditions of both symmetric and asymmetric information. The research compares the effects of full-commitment contracts and anti-renegotiation contracts on the strategies of brand owners, recyclers, and information incentives.

Determining how children solve arithmetic problems when they stop using their fingers is a real challenge. To take it up, the evolution of problem-size effects for tie and non-tie problems was observed when 6-year-olds (N = 65) shift from finger counting to mental strategies. These observations revealed that the problem-size effect remained the same for non-tie problems, whereas it drastically decreased for tie problems.

Background: Duchenne muscular dystrophy (DMD) is a devastating disease characterized by progressive muscle wasting that leads to diminished lifespan. In addition to the inherent weakness of dystrophin-deficient muscle, the dysfunction of resident muscle stem cells (MuSC) significantly contributes to disease progression.
Methods: Using the mdx mouse model of DMD, we performed an in-depth characterization of disease progression and MuSC function in dystrophin-deficient skeletal muscle using immunohistology, isometric force measurements, transcriptomic analysis and transplantation assays.
