Bulk gene expression experiments relied on aggregations of thousands of cells to measure the average expression in an organism. Advances in microfluidic and droplet sequencing now permit expression profiling in single cells. This study of cell-to-cell variation reveals that individual cells lack detectable expression of transcripts that appear abundant on a population level, giving rise to zero-inflated expression patterns. To infer gene co-regulatory networks from such data, we propose a multivariate Hurdle model. It is comprised of a mixture of singular Gaussian distributions. We employ neighborhood selection with the pseudo-likelihood and a group lasso penalty to select and fit undirected graphical models that capture conditional independences between genes. The proposed method is more sensitive than existing approaches in simulations, even under departures from our Hurdle model. The method is applied to data for T follicular helper cells, and a high-dimensional profile of mouse dendritic cells. It infers network structure not revealed by other methods; or in bulk data sets. Objectives: Although the multifactorial nature of chronic pain is well established, research has predominantly examined isolated variables or singular pathways that may contribute to this condition. We use a complex systems perspective to examine the interplay of psychological factors in the context of chronic pain.
Methods: We analyzed two cross-sectional datasets (N=935 and 1366) collected at a pain clinic and rehabilitation center in Belgium from individuals primarily with musculoskeletal pain.

This research explores the dynamical properties and solutions of actin filaments, which serve as electrical conduits for ion transport along their lengths. Utilizing the Lie symmetry approach, we identify symmetry reductions that simplify the governing equation by lowering its dimensionality. This process leads to the formulation of a second-order differential equation, which, upon applying a Galilean transformation, is further converted into a system of first-order differential equations.

Objectives: The National Institute for Health and Care Excellence (NICE) in England introduced early value assessments (EVAs) as an evidence-based method of accelerating access to promising health technologies that could address unmet needs and contribute to the National Health Service's Long Term Plan. However, there are currently no published works considering differences and commonalities in methods used between Assessment Reports for EVAs.
Methods: This rapid scoping review included all completed EVAs published on the NICE website up to 23 July 2024.
