Article Abstract

The formation of host-guest complexes between seven flavylium cations and water-soluble -sulfonatocalix[4]arene (SC4) was investigated by UV/vis absorption, fluorescence, and NMR spectroscopies. The results show the cationic guests form complexes with affinities in the submillimolar range. A representative chalcone/flavylium photoswitch was investigated in more detail regarding its pH- and light-triggered interconversion between the two forms. The dramatic affinity differentiation of the SC4 binding of the two switchable species (40 M for the -chalcone versus 3.5 × 10 M for the flavylium cation) enables the pH-gated photocontrol of the complexation process. These responsive properties were explored to demonstrate the competitive and selective release of biologically relevant guests from their supramolecular complexes with -sulfonatocalix[4]arene (SC4), following the principle of AND logic. The guest release can be reverted by the thermally activated reaction of the flavylium ion back to the -chalcone.

The E76K mutation in protein tyrosine phosphatase (PTP) SHP2 is a recurrent driver of developmental disorders and cancers, yet the mechanism by which this single-site substitution promotes persistent activation remains elusive. Here, we combine path-based conformational sampling, unbiased molecular dynamics (MD) simulations, Markov state models (MSMs), and neural relational inference (NRI) to elucidate how E76K reshapes the activation landscape and regulatory architecture of SHP2. Advances in data collection have resulted in an exponential growth of high-dimensional microarray datasets for binary classification in bioinformatics and medical diagnostics. These datasets generally possess many features but relatively few samples, resulting in challenges associated with the "curse of dimensionality", such as feature redundancy and an elevated risk of overfitting. While traditional feature selection approaches, such as filter-based and wrapper-based approaches, can help to reduce dimensionality, they often struggle to capture feature interactions while adequately preserving model generalization.
