A series of tetracationic quadrupolar chromophores containing three-coordinate boron π-acceptors linked by different π-bridges, namely 4,4'-biphenyl, 2,7-pyrene, 2,7-fluorene, 3,6-carbazole and 5,5'-di(thien-2-yl)-3,6-diketopyrrolopyrrole, were synthesized. While their neutral precursors displayed highly solvatochromic fluorescence, the water-soluble tetracationic target molecules , did not, but their emission colour could be tuned from blue to pink by changing the π-bridge. Compound , containing the diketopyrrolopyrrole bridge, exhibits the most red-shifted absorption and emission maxima and the largest two-photon absorption cross-section (4560 GM at 740 nm in MeCN). Confocal laser scanning fluorescence microscopy studies in live cells confirm localization of the dye at the lysosome. Moreover, the low cytotoxicity, and high photostability of combined with two-photon excited fluorescence imaging studies demonstrate its excellent potential for lysosomal imaging in live cells.

Epithelial cells work collectively to provide a protective barrier, yet they turn over rapidly through cell division and death. If the numbers of dividing and dying cells do not match, the barrier can vanish, or tumours can form. Mechanical forces through the stretch-activated ion channel Piezo1 link both of the processes; stretch promotes cell division, whereas crowding triggers live cells to extrude and then die.

A practical and novel technique used both in testing and in practice when tightening dental implant screw systems is described for determining the preload implant screw systems, which differs from traditional tightening procedures that are based solely on the application of a predetermined manufacturer specified torque value. Preload is the critical quantity for a reliable joint: about 90% of tightening torque goes into overcoming friction and the remaining amount, approximately 10%, goes into preload. Because of the heavy dependence of torque on friction, the actual preload achieved is subject to large variability of up ±35%, depending on surface conditions (dry, wet, or contaminated).

Glycine is an important metabolite and cell signal in diverse organisms, yet tools to visualize intracellular glycine dynamics have not been developed. In this study, diverse and bright RNA-based glycine biosensors were developed by fusing the architecturally complex glycine riboswitch with Broccoli class fluorogenic aptamers. The brightest sensor with the highest activation, glyS, and its two-dye ratiometric counterpart, Pepper-glyS, allowed for visualization of a drug-induced accumulation of endogenous glycine in live Escherichia colicells.

Fetal bovine serum (FBS) is an undefined additive that is ubiquitous to mammalian cell culture media and whose functional contributions to promoting cell proliferation remain poorly understood. Efforts to replace serum supplementation in culture media have been hindered by an incomplete understanding of the environmental requirements fulfilled by FBS. Here, we use a combination of live-cell imaging and quantitative lipidomics to elucidate the role of serum in supporting proliferation.

Lineage specification requires accurate interpretation of multiple signaling cues. However, how combinatorial signaling histories influence fate outcomes remains unclear. We combined single-cell transcriptomics, live-cell imaging, and mathematical modeling to explore how activin and bone morphogenetic protein 4 (BMP4) guide fate specification during human gastrulation.
