Article Abstract

Lysosomes are acidic organelles responsible for degrading both exogenous and endogenous materials. The small GTPase Arl8 localizes primarily to lysosomes and is involved in lysosomal function. In the present study, using Arl8b gene-trapped mutant (Arl8b ) mice, we show that Arl8b is required for the development of dorsal structures of the neural tube, including the thalamus and hippocampus. In embryonic day (E) 10.5 Arl8b embryos, Sox1 (a neuroepithelium marker) was ectopically expressed in the roof plate, whereas the expression of Gdf7 and Msx1 (roof plate markers) was reduced in the dorsal midline of the midbrain. Ectopic expression of Sox1 in Arl8b embryos was detected also at E9.0 in the neural fold, which gives rise to the roof plate. In addition, the levels of Bmp receptor IA and phosphorylated Smad 1/5/8 (downstream of BMP signaling) were increased in the neural fold of E9.0 Arl8b embryos. These results suggest that Arl8b is involved in the development of the neural fold and the subsequently formed roof plate, possibly via control of BMP signaling.

Apical constriction of epithelial cells usually occurs in a local portion of epithelial sheet, which results in bending of epithelial tissues. However, it is uncertain whether diffusible signal molecules, like Wnt, regulate such locally restricted events. Here, we show that Wnt ligands are required for apical constriction of Wnt1-expressing roof plate (RP) cells during development of the neural tube.

Mastering the deformation and air leakage patterns of gob-side retained roadway in fully mechanized mining faces, as well as the distribution characteristics of the gas flow field, is of great significance for the comprehensive prevention and control of gas in the goaf. Taking the 20,107 fully mechanized mining face of Qincheng Coal Mine in Shanxi Province as a case study, this paper employs fracture mechanics and plate-beam theory to analyze the impact of the roof fracture position on stress concentration and deformation failure of the roadway wall. It is found that when the fracture line is located outside the wall, the supporting stress can be effectively transferred and pressure can be relieved.

The current study aims to compare the morphological features of the oropharyngeal roof of the little owl (Athene noctua) and the domestic pigeon (Columba livia domestica) by gross anatomy, morphometric measurements, scanning electron microscopy, and light microscopy. Ten heads of normal, healthy, and adult little owls and domestic pigeons were used in the current study as follows: seven heads for gross anatomical examination and light microscopy, and three for scanning electron microscopy. The oropharyngeal roof had a triangular appearance in both birds, was enlarged and broad in the little owl, and narrowly elongated in the domestic pigeon.

However, they rapidly dedifferentiate in vitro, challenging large-scale maintenance.
