Article Abstract

Aberrant mitochondrial dynamics disrupts mitochondrial function and contributes to disease conditions. A targeted RNA interference screen for deubiquitinating enzymes (DUBs) affecting protein levels of multifunctional mitochondrial fusion protein Mitofusin (MFN) identified USP8 prominently influencing MFN levels. Genetic and pharmacological inhibition of USP8 normalized the elevated MFN protein levels observed in PINK1 and Parkin-deficient models. This correlated with improved mitochondrial function, locomotor performance and life span, and prevented dopaminergic neurons loss in PINK1 KO flies. We identified a novel target antagonizing pathologically elevated MFN levels, mitochondrial dysfunction, and dopaminergic neuron loss of a model of mitochondrial dysfunction.

Mutations in the human ADAR gene encoding adenosine deaminase acting on RNA 1 (ADAR1) cause Aicardi-Goutières syndrome 6 (AGS6); a severe auto-inflammatory encephalopathy with aberrant interferon (IFN) induction. AdarΔ2-13 null mutant mouse embryos lacking ADAR1 protein die with high levels of IFN-stimulated gene (ISG) transcripts. In Adar Mavs double mutants also lacking the Mitochondrial antiviral signaling (MAVS) adaptor, the aberrant IFN induction is prevented.

Hyperglycemia-induced oxidative stress and inflammation critically impair diabetic bone defect repair. Here, a radially oriented microchannel scaffold (D-GSH@QZ) was developed via a directional freezing technique integrated with photo-cross-linking strategies. The family Syngnathidae includes seahorses, sea dragons, and pipefishes. We sequenced the complete mitochondrial DNA (mtDNA) genome of the belly pipefish, Bleeker, 1849. The genome is 16,646-bp long, and includes the standard complement for bony fishes of 13 protein-coding genes, 22 tRNA genes, two rRNA genes, and a control region, in the same order and strand distribution as other syngnathids.
