Article Abstract

Protein nanocapsules are potentially useful as functional nanocarriers because of their hollow structure and high biocompatibility and the intrinsic activity of their protein constituents. However, the development of a facile method for the preparation of oriented nanocapsules that retain their protein activity has been challenging. Here we describe the preparation of protein nanocapsules through the enzymatic removal of polymer templates. Nickel(II) nitrilotriacetic acid-end-functionalized poly(lactic acid) (Ni-NTA-PLA) was introduced as a polymeric template to immobilize hexa-histidine-tagged green fluorescence protein (His-GFP) with consistent orientation. Following protein cross-linking and core-degradation, various measurements as a function of degradation time indicated the formation of hollow structures. We also demonstrated orientational control and activity preservation of the protein after capsule preparation. Protein nanocapsules prepared by this method can act as functional containers, taking advantage of the intrinsic function of their constituent proteins without additional modification.

Cancer constitutes a major cause of death globally. Many current treatments are not very selective and often harm healthy cells. Inflammation is known to be associated with tumor growth, yet anti-inflammatory drugs alone are rarely used in a targeted manner.

Introduction: Traumatic brain injury is the leading cause of death and disability in individuals under 40 years old. It induces various neuropathological outcomes, including cognitive, emotional, and physiological deficits, likely linked to early neuroinflammatory processes. In an animal model, mild traumatic brain injury (mTBI) has been shown to elevate oxidative stress biomarkers, such as advanced oxidation protein products (AOPP) and malondialdehyde (MDA), which persist for over a week.

Dysregulation of pro-inflammatory cytokines participates in the initiation and development of knee osteoarthritis (OA). Consequently, interventions to boost the anti-inflammatory capacity of articular chondrocytes have been proposed to treat early-stage OA and prevent OA progression. Applying nanoencapsulation can enhance bioavailability and bioactivity and sustain the anti-inflammatory activity of phytochemicals.

Global concerns about environmental pollution, poor waste management, and the rise in antimicrobial resistance due to uncontrolled antibiotic use have driven researchers to seek alternative, multifaceted solutions. Plants, animals, microorganisms, and their processing wastes serve as valuable sources of natural biopolymers and bioactive compounds. Through nanotechnology, these can be assembled into formulations with enhanced antimicrobial properties, high safety, and low toxicity.
