The synthesis of three new cyclometallated platinum(ii) compounds containing a para-tolyl ligand and a tridentate [C,N,N'] (cm1) or a bidentate [C,N] ligand and an additional ligand such as SEt2 (cm2) or PPh3 (cm3) is reported. The X-ray molecular structure of platinum(ii) compound cm3 is also presented. Intermolecular oxidative addition of methyl iodide or iodine upon cm1, cm2 and cm3 produced six novel cyclometallated platinum(iv) compounds. The cytotoxic activity against a panel of human adenocarcinoma cell lines (A-549 lung, MDA-MB-231 and MCF-7 breast, and HCT-116 colon), DNA interaction, topoisomerase I, IIα, and cathepsin B inhibition, and cell cycle arrest, apoptosis and ROS generation of the investigated complexes are presented. The best results for antiproliferative activity were obtained for platinum(iv) compounds cm1MeI and cm1I2 arising from oxidative addition of methyl iodide and iodine, respectively, to cm1. Cyclometallated platinum(iv) compounds cm1MeI and cm3MeI induce significant changes in the mobility of DNA and, in addition, cm1MeI, cm3MeI and cm1I2, showed considerable topoisomerase IIα inhibitory activity. Moreover, the compounds exhibiting the higher antiproliferative activity (cm1MeI and cm1I2) were found to generate ROS and to supress HCT-116 colon cancer cell growth by a mixture of cell cycle arrest and apoptosis induction. 1H NMR experiments carried out in a buffered aqueous medium (pH 7.40) indicate that compound cm1MeI is not reduced by common biologically relevant reducing agents such as ascorbic acid, glutathione or cysteine.

Dicationic, -symmetrical, tris-chelate Pt(IV) complexes of general formula [Pt(trz)(N∧N)](OTf), bearing two cyclometalated 4-butyl-3-methyl-1-phenyl-1-1,2,3-triazol-5-ylidene (trz) ligands and one aromatic diimine [N∧N = 2,2'-bipyridine (bpy, ), 4,4'-di--butyl-2,2'-bipyridine (dbbpy, ), 4,4'-dimethoxi-2,2'-bipyridine (dMeO-bpy, ), 1,10-phenanthroline (phen, ), 4,7-diphenyl-1,10-phenanthroline (bphen, ), dipyrido[3,2-:2',3'-]phenazine (dppz, ), or 2,3-diphenylpyrazino[2,3-][1,10]phenanthroline (dpprzphen, )] are obtained through chloride abstraction from [PtCl(trz)] () using AgOTf in the presence of the corresponding diimine. Platinum(II) complexes of -cyclometalating ligands based on 6-phenyl-2,2'-bipyridine (HL) have been widely investigated for their luminescence properties. We describe how PtLCl and five analogues with differently substituted aryl rings, PtLCl, can be oxidized with chlorine and/or iodobenzene dichloride to generate Pt(IV) compounds of the form Pt(-L)Cl ( = 1-6). The molecular structures of several of them have been determined by X-ray diffraction.

The first series of neutral, tris-chelate, phosphorecent Pt(IV) complexes is reported, which combine two cyclometalated 2-arylpyridine ligands and a dimetalated biaryl. The introduction of biaryl ligands is achieved under mild conditions through the oxidative addition of dibenzoiodolium ions to Pt(II) precursors to give Pt(IV) intermediates with a singly metalated 2-(2-iodoaryl)aryl ligand, followed by the reductive metalation of the C-I bond. The modulation of emission characteristics derivatization of both types of ligands is demonstrated.

We present a Pt-catalyzed direct coupling of benzene to biphenyl. This catalytic reaction employs a cyclometalated platinum(ii) complex [PtMe(bhq)(SMe2)] (bhq = benzo[h]quinolate) with PhI(OAc)2 as an oxidant and does not require an acid, a co-catalyst or a solvent. The reaction kinetics and characterization of potential catalytic species are reported.
