In Huntington's disease (HD), expansion of CAG codons in the huntingtin gene (HTT) leads to the aberrant formation of protein aggregates and the differential degeneration of striatal medium spiny neurons (MSNs). Modeling HD using patient-specific MSNs has been challenging, as neurons differentiated from induced pluripotent stem cells are free of aggregates and lack an overt cell death phenotype. Here we generated MSNs from HD patient fibroblasts through microRNA-based direct neuronal conversion, bypassing the induction of pluripotency and retaining age signatures of the original fibroblasts. We found that patient MSNs consistently exhibited mutant HTT (mHTT) aggregates, mHTT-dependent DNA damage, mitochondrial dysfunction and spontaneous degeneration in culture over time. We further provide evidence that erasure of age stored in starting fibroblasts or neuronal conversion of presymptomatic HD patient fibroblasts results in differential manifestation of cellular phenotypes associated with HD, highlighting the importance of age in modeling late-onset neurological disorders.

Orthodontic-induced gingival enlargement (OIGE) affects approximately 15-30% of patients undergoing orthodontic treatment and remains largely unpredictable, often relying on subjective clinical assessments made after irreversible tissue changes have occurred. S100A4 is a well-characterized marker of activated fibroblasts involved in pathological tissue remodeling. This was a cross-sectional precision biomarker study that analyzed gingival tissue samples from three groups: healthy controls (n = 60), orthodontic patients without gingival enlargement (n = 31), and patients with clinically diagnosed OIGE (n = 61).

Solitary fibrous tumor, formerly also referred to as hemangiopericytoma, is a fibroblastic mesenchymal neoplasm of the orbit that rarely metastasizes but has a propensity for local recurrence. We describe 5 cases of orbital solitary fibrous tumors that were treated with radiation. In 3 patients, radiation therapy was given in the neoadjuvant setting prior to surgery.

Purpose: In this retrospective study, whether [Ga]Ga-DOTA-FAPI-04 PET/MR imaging biomarkers can predict the progression-free survival (PFS) and overall survival (OS) of patients with advanced pancreatic cancer was investigated.
Methods: Fifty-one patients who underwent [Ga]Ga-DOTA-FAPI-04 PET/MR scans before first-line chemotherapy were recruited. Imaging biomarkers, including the maximum tumor diameter, minimum apparent diffusion coefficient (ADC), maximum and mean standardized uptake values (SUV and SUV), fibroblast activation protein- (FAP-) positive tumor volume (FTV and W-FTV) and total lesion FAP expression (TLF and W-TLF), were recorded for primary and whole-body tumors.
