We describe investigations to expand the scope of next generation maleimide cross-linkers for the construction of homogeneous protein-protein conjugates. Diiodomaleimides are shown to offer the ideal properties of rapid bioconjugation with reduced hydrolysis, allowing the cross-linking of even sterically hindered systems. The optimized linkers are exploited to link human serum albumin to antibody fragments (Fab or scFv) as a prospective half-life extension platform, with retention of antigen binding and robust serum stability. Finally, a triprotein conjugate is formed, by linking scFv antibody fragments targeting carcinoembryonic antigen. This tri-scFv is shown to infer a combination of greater antigen avidity and increased in vivo half-life, representing a promising platform for antibody therapeutic development.

Group II introns are self-splicing ribozymes that excise themselves from precursor RNA and integrate into new DNA locations through retromobility. Splicing is facilitated by an intron-encoded protein (IEP), a multidomain reverse transcriptase that enhances ribozyme activity and promotes formation of lariat intron-IEP ribonucleoprotein (RNP) complexes. In this study, we examined the role of conserved cysteine residues in the IEP of the group IIC intron I1 from the thermophile by generating cysteine-to-methionine mutants.

Here, in the present article, a novel molecularly imprinted polymer (MIP), S-RV-P, was prepared for the recognition and separation of the S-enantiomer of rivastigmine (S-RV) from its racemic form. Synthesis involved the formation of a poly(styrene-co-maleic anhydride) (PSM) matrix that was functionalized with 2-aminoethane-1-thiol (ATH) to introduce thiol and carboxylic acid functionalities to prepare the precursor HS-P-COOH. Post-imprinting was achieved through thiol-maleimide click crosslinking via bismaleimide (BMA) facilitated by S-RV to generate the resultant S-RV-P polymer.

TUB-010 is a next-generation antibody-drug conjugate (ADC) targeting CD30 expressed on various hematopoietic malignancies such as Hodgkin lymphoma. Among the therapeutic options for patients with relapsed and refractory CD30-positive cancers is brentuximab vedotin (Adcetris), an MMAE-delivering anti-CD30 ADC with a mean drug-to antibody ratio (DAR) of 4. Adcetris exhibits a high response rate at the cost of significant toxicities, likely driven by the payload MMAE and instability of the maleimide conjugation chemistry.

Customized solid-state lithium metal batteries (SSLMBs) with high safety hold promise for next-generation energy storage systems, yet they suffer from rapid capacity decay due to unstable solid-solid interfaces and uneven Li transport. To break the bottleneck, an in situ 3D-printed integrated porous cathode/composite polymer electrolyte (CPE) is developed using reversible Diels-Alder (DA) covalent chemistry. Typically, the reversible crosslinked CPE are produced from a furan-functionalized polyethylene glycol oligomer, tris-(2-maleimidoethyl)amine, and maleimide-modified LiLaZrTaO.
