Article Abstract

Dynamic imaging analyses of antigen-specific T-B interactions in germinal centers have advanced our understanding of the molecular mechanisms underlying affinity maturation and provided a wealth of information about how follicular helper T cells function in vivo. Here we describe a routine method to visualize fluorescence protein-expressing, antigen-specific T and B cells in germinal centers. The protocol for incorporating functional reporters or genetic perturbation of the T cells by retroviral transduction is also briefly described, using the FRET-based calcium reporter as an example.

The fraction that the elderly represent in the world's population is growing rapidly; numerous alterations that impact all organs and systems, including the immune system, are related to aging. A complex process common in the elderly, known as immunosenescence, is characterized by a decreased ability to respond to vaccination as well as an increased risk of bacterial and viral infections, autoimmune, cardiovascular and neurodegenerative diseases. These processes are associated with alterations in the innate and adaptive immune system and lead to a condition of chronic low-grade inflammation, referred to as inflammaging.

To better understand the contribution of interferon-γ (IFN-γ) receptor signaling to vaccine-induced immunity, we employed A129 (IFN-α/β receptor-deficient) and AG129 (IFN-α/β/γ receptor-deficient) mouse models. AG129 mice induced comparable levels of virus-specific IgG after vaccination with influenza virus H5 hemagglutinin (HA) virus-like particles (VLPs). Vaccinated AG129 mice with HA VLPs exhibited impaired Th1-immune responses, lower hemagglutination inhibition (HAI) titers, increased susceptibility to virus infection, and lower survival rates following influenza virus (H5N1) challenge than vaccinated A129 mice.

Castleman disease (CD) is a rare lymphoproliferative disorder with unique clinicopathological features, including two distinct clinical subtypes categorized as unicentric (UCD) and multicentric (MCD). UCD usually involves a single lymph node site presenting with no or minimal local symptoms. Histologically, most UCD cases exhibit regressive hyaline vascular germinal centers, characterized by penetrating vessels, dendritic hyperplasia/dysplasia, and increased interfollicular vascularity.
