The significance of Cdk5 in cell-cycle control and cancer biology has gained increased attention. Here we report the inverse correlation between the protein levels of Cdk5 and p21 from cell-based and clinical analysis. Mechanistically, we identify that Cdk5 overexpression triggers the proteasome-dependent degradation of p21 through a S130 phosphorylation in a Cdk2-independent manner. Besides, the evidence from cell-based and clinical analysis shows that Cdk5 primarily regulates nuclear p21 protein degradation. S130A-p21 mutant enables to block either its protein degradation or the increase of cancer cell growth caused by Cdk5. Notably, Cdk5-triggered p21 targeting primarily appears in S-phase, while Cdk5 overexpression increases the activation of Cdk2 and its interaction with DNA polymerase δ. The in vivo results show that Cdk2 might play an important role in the downstream signaling to Cdk5. In summary, these findings suggest that Cdk5 in a high expression status promotes cancer growth by directly and rapidly releasing p21-dependent cell-cycle inhibition and subsequent Cdk2 activation, which illustrates an oncogenic role of Cdk5 potentially applied for future diagnosis and therapy. Neurons that encode odor information are fundamental to innate fear processes, yet how mitral/tufted (M/T) cells encode innate fear remains unknown. Here, we identify three different response patterns of M/T cells in the dorsal olfactory bulb (dOB) during active avoidance elicited by non-dehydrogenated 2,4,5-trimethylthiazole (nTMT) through in vivo calcium imaging and multielectrode recordings in mice, including enhanced responses, suppressed responses, and no response. Remarkably, suppressed response M/T cells encode active avoidance, whereas suppressed and enhanced response M/T cells jointly encode passive freezing.

Two series of triazolo[1,5-a]pyrimidines were designed and synthesized as antiproliferative agents targeting multi kinase inhibition aiming to increase potency and combat drug resistance. The synthesized compounds were tested for their antiproliferative activity. The triazolopyrimidine derivatives 9b, 9c, 12b and 12c showed promising anticancer activities, in particular, compounds 12b and 12c displayed broad spectrum antiproliferative potential against NCI cancer cell lines with GI mean value of 10.

Parkinson's disease (PD) is characterized by the loss of dopaminergic neurons in the substantia nigra and the presence of α-synuclein-positive inclusions known as Lewy bodies. Synphilin-1 is a protein of unknown function that interacts with α-synuclein and has been shown to exhibit cytoprotective effects in both in vitro and in vivo models.
