HIV-1-infected individuals harbor a latent reservoir of infected CD4 T cells that is not eradicated by antiretroviral therapy (ART). This reservoir presents the greatest barrier to an HIV-1 cure and has remained difficult to characterize, in part, because the vast majority of integrated sequences are defective and incapable of reactivation. To characterize the replication-competent reservoir, we have combined two techniques, quantitative viral outgrowth and qualitative sequence analysis of clonal outgrowth viruses. Leukapheresis samples from four fully ART-suppressed, chronically infected individuals were assayed at two time points separated by a 4- to 6-mo interval. Overall, 54% of the viruses emerging from the latent reservoir showed gp160 env sequences that were identical to at least one other virus. Moreover, 43% of the env sequences from viruses emerging from the reservoir were part of identical groups at the two time points. Groups of identical expanded sequences made up 54% of proviral DNA, and, as might be expected, the sequences of replication-competent viruses in the active reservoir showed limited overlap with integrated proviral DNA, most of which is known to represent defective viruses. Finally, there was an inverse correlation between proviral DNA clone size and the probability of reactivation, suggesting that replication-competent viruses are less likely to be found among highly expanded provirus-containing cell clones.

Within these RCs, viral genome replication, gene expression, and modulation of cellular antiviral responses are tightly coordinated, making them valuable models for studying virus-host interactions. In a recent study, we analyzed the protein composition of HAdV type 5 (HAdV-C5) RCs isolated from infected primary cells at different time points during infection using quantitative proteomics.

Introduction: Low-level viremia (LLV) in HIV infection, defined as detectable but low plasma viral load, is associated with an increased risk of virological failure (VF); however, the mechanisms underlying LLV remain unclear. Monocytes, as potential viral reservoirs, can migrate into tissues and differentiate into tissue-resident macrophage reservoirs, playing a critical role in viral dissemination and potentially driving persistent viremia.

ObjectivesHistorically, vaccines have been administered in the dorsal interscapular region of cats (the 'scruff' of the neck) owing to easy access to the subcutaneous space. In response to concerns about sarcomas developing at injection sites (feline injection site sarcomas [FISSs]), and a possible association between feline leukaemia virus (FeLV) vaccination and the development of FISS, alternative FeLV vaccination sites such as the distal left hindlimb and tail have been proposed by influential vaccination bodies and various key opinion leaders. There is a dearth of evidence, however, to demonstrate the development of a comparable immune response after FeLV vaccination in these sites.

Antibodies to programmed cell death 1 (PD-1), Programmed death-ligand 1 (PDL-1) and Cytotoxic-T-lymphocyte-associated protein 4 (CTLA-4) can revert HIV latency and enhance anti-HIV cytotoxic response but their impact on HIV proviral sequences and integration landscape in people with HIV (PWH) remain to be studied. Two PWH treated with PD-1/PDL-1 and one with PD-1/CTLA4 were studied among the ANRS-CO-24 OncoVIHAC cohort study. Matched integration site and proviral sequencing were performed pre- and post-treatment.

Porcine circovirus type 2 (PCV2) is a globally prevalent swine pathogen that induces immunosuppression, predisposing pigs to subclinical infections. In intensive farming systems, PCV2 persistently impairs growth performance and vaccine efficacy, leading to substantial economic losses in the swine industry. Emerging evidence suggests that certain viruses exploit Suppressor of Cytokine Signaling 3 (SOCS3), a key immune checkpoint protein, to subvert host innate immunity by suppressing cytokine signaling.
