Article Abstract

We report a tool for the analysis of subcellular proteomics data, called MetaMass, based on the use of standardized lists of subcellular markers. We analyzed data from 11 studies using MetaMass, mapping the subcellular location of 5,970 proteins. Our analysis revealed large variations in the performance of subcellular fractionation protocols as well as systematic biases in protein annotation databases. The Excel and R versions of MetaMass should enhance transparency and reproducibility in subcellular proteomics.

Megalencephalic leukoencephalopathy with subcortical cysts (MLC) is a rare leukodystrophy characterized by early-onset macrocephaly, white matter edema, seizures, and motor and cognitive decline. Missense mutations to hepatic and glial cell adhesion molecule (hepaCAM), also known as GlialCAM, are responsible for approximately twenty-five percent of MLC cases. HepaCAM is highly enriched in astrocytes and plays important roles in astrocyte territory establishment, gap junction coupling, branching organization, synaptic function, and development of the gliovascular unit.

Single-cell proteomics by mass spectrometry is advancing rapidly, yet throughput and sensitivity remain limiting-particularly for small, protein-poor cell types such as neutrophils. As the most abundant circulating leukocytes in humans, neutrophils are central to immune defense and inflammation, but their proteomes comprehensive single-cell level characterization has only concurrently been reported and remains limited. Here, we introduce a rapid capillary electrophoresis-mass spectrometry (CE-MS) platform, which integrates electrophoresis-correlative real-time data acquisition with sub-7-minute separations and artificial intelligence (AI)-based data processing software to achieve deep, high-throughput profiling.
