Article Abstract

Polycomb Group (PcG) proteins are epigenetic repressors essential for control of development and cell differentiation. They form multiple complexes of which PRC1 and PRC2 are evolutionary conserved and obligatory for repression. The targeting of PRC1 and PRC2 is poorly understood and was proposed to be hierarchical and involve tri-methylation of histone H3 (H3K27me3) and/or monoubiquitylation of histone H2A (H2AK118ub). Here, we present a strict test of this hypothesis using the Drosophila model. We discover that neither H3K27me3 nor H2AK118ub is required for targeting PRC complexes to Polycomb Response Elements (PREs). We find that PRC1 can bind PREs in the absence of PRC2 but at many PREs PRC2 requires PRC1 to be targeted. We show that one role of H3K27me3 is to allow PcG complexes anchored at PREs to interact with surrounding chromatin. In contrast, the bulk of H2AK118ub is unrelated to PcG repression. These findings radically change our view of how PcG repression is targeted and suggest that PRC1 and PRC2 can communicate independently of histone modifications.

Polycomb group (PcG) proteins are repressors of developmental genes. Paradoxically, the same PcG proteins also function in gene activation via mechanisms that are not yet fully understood. Here, we found that SKP1A, an essential factor of SKP1A/CUL1/F-box (SCF) ubiquitin ligases and Polycomb-repressive complex 1 (PRC1) containing PCGF1 (PCGF1-PRC1), mediates the link between PcG-dependent gene regulation and ubiquitin proteasomal degradation.

Heterozygous missense mutations in cause Weaver syndrome (WS), a developmental disorder characterized by intellectual disability and overgrowth. encodes the enzymatic subunit of Polycomb repressive complex 2 (PRC2), which mediates monomethylation, dimethylation, and trimethylation of histone H3 lysine 27 (H3K27me1/2/3). Most WS-associated EZH2 variants lack functional characterization but are presumed loss-of-function.

The Polycomb system epigenetically represses selected developmental genes to enforce gene expression programs of differentiated cells. The system requires the coordinated action of dozens of structurally unrelated proteins assembled in two evolutionarily conserved polycomb repressive complexes, PRC1 and PRC2. Genes repressed by the Polycomb system are enriched in histone H3 trimethylated at lysine 27 (H3K27me3), an epigenetic mark that propagates the repressed state after DNA replication.

Polycomb repressive complexes (PRC) and nucleosome remodeling and deacetylase (NuRD) complex are crucial for regulating the expression of pluripotent and developmental genes and maintaining the characteristics of mouse embryonic stem cells (mESCs). However, the interplay between the Polycomb and NuRD complexes in mESCs, particularly under protein kinase C (PKC) inhibition, remains to be elucidated. We knocked down Polycomb complexes components Ezh2, Ring1b, and Cbx7 via short hairpin RNA interference and observed significant reductions in most NuRD complex components, especially Mbd3, Mta1, Rbbp4, and Rbbp7.
