Mycothiol serves as the primary reducing agent in Mycobacterium species, and is also a cofactor for the detoxification of xenobiotics. Mycothiol conjugate amidase (Mca) is a metalloamidase that catalyzes the cleavage of MS-conjugates to form a mercapturic acid, which is excreted from the mycobacterium, and 1-D-myo-inosityl-2-amino-2-deoxy-α-D-glucopyranoside. Herein we report on the metal cofactor preferences of Mca from Mycobacterium smegmatis and Mycobacterium tuberculosis. Importantly, results from homology models of Mca from M. smegmatis and M. tuberculosis suggest that the metal binding site of Mca is identical to that of the closely related protein N-acetyl-1-D-myo-inosityl-2-amino-2-deoxy-α-D-glucopyranoside deacetylase (MshB). This finding is supported by results from zinc ion affinity measurements that indicate Mca and MshB have comparable K(D)(ZnII) values (~10-20 pM). Furthermore, results from pull-down experiments using Halo-Mca indicate that Mca purifies with (stoichiometric) Fe(2+) when purified under anaerobic conditions, and Zn(2+) when purified under aerobic conditions. Consequently, Mca is likely a Fe(2+)-dependent enzyme under physiological conditions; with Zn(2+)-Mca an experimental artifact that could become biologically relevant under oxidatively stressed conditions. Importantly, these findings suggest that efforts towards the design of Mca inhibitors should include targeting the Fe(2+) form of the enzyme.

We report a 64-year-old woman who developed symptomatic vasospasm on postoperative day 7 after clipping of an unruptured right middle cerebral artery (MCA) aneurysm. Imaging revealed right MCA vasospasm, which resolved with oral antiplatelets and intravenous vasodilators. She was discharged without neurological deficits.

Clonal hematopoiesis of indeterminate potential (CHIP) bridges hematopoietic clonality and solid tumorigenesis, unveiling a systemic dimension of somatic mutagenesis in cancer biology. Generally, population studies demonstrate CHIP carriers face elevated risks of cancer and poorer survival outcomes. This review consolidates current knowledge on the role of CHIP as potential biomarkers in the prediction/early detection/prognosis evaluation of various non-hematological cancers.

The onset of puberty is increasingly observed at earlier ages in children, especially in girls with obesity, a trend that predisposes them to long-term metabolic and reproductive disorders in adulthood. Bile acids have emerged as pivotal signaling molecules in both metabolic and reproductive disorders, but remain unexplored in the early onset of puberty in children. Herein, we find elevated levels of muricholic acid (MCA) species in the serum of girls with central precocious puberty, which strongly correlate with indices of hypothalamic-pituitary-gonadal axis activation and can reach peak levels during puberty among healthy children.

Large vessel occlusion (LVO)-induced acute ischemic stroke (AIS) manifests with abrupt onset and critical severity, primarily caused by cerebral artery atherosclerosis or cardiogenic embolism leading to luminal stenosis or occlusion. The authors report a case of acute massive cerebral infarction caused by occlusion of the left middle cerebral artery (MCA) and missed the time window for thrombolysis, in which spontaneous recanalization of the MCA was achieved after 1 week of conservative treatment. After 2 months of rehabilitation training, neurological functions and other symptoms significantly improved.
