Article Abstract

Secondary structure formation in oligopeptides can be induced by short nucleating segments with a high propensity to form hydrogen bonded turn conformations. Type I/III turns facilitate helical folding while type II'/I' turns favour hairpin formation. This principle is experimentally verified by studies of two designed dodecapeptides, Boc-Val-Phe-Leu-Phe-Val-Aib-Aib-Val-Phe-Leu-Phe-Val-OMe 1 and Boc-Val-Phe-Leu-Phe-Val-(D)Pro-(L)Pro-Val-Phe-Leu-Phe-Val-OMe 2. The N- and C-terminal flanking pentapeptide sequences in both cases are identical. Peptide 1 adopts a largely α-helical conformation in crystals, with a small 310 helical segment at the N-terminus. The overall helical fold is maintained in methanol solution as evidenced by NMR studies. Peptide 2 adopts an antiparallel β-hairpin conformation stabilized by 6 interstrand hydrogen bonds. Key nuclear Overhauser effects (NOEs) provide evidence for the antiparallel β-hairpin structure. Aromatic proton chemical shifts provide a clear distinction between the conformation of peptides 1 (helical) and 2 (β-hairpin). The proximity of facing aromatic residues positioned at non-hydrogen bonding positions in the hairpin results in extensively ring current shifted proton resonances in peptide 2.

The abnormal expansion of GGGGCC (G4C2) repeats in the noncoding region of the C9orf72 gene is a major genetic cause of two devastating neurodegenerative disorders, amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). These G4C2 repeats are known to form G-quadruplex (G4) structures, which are hypothesized to contribute to disease pathogenesis. Here, we demonstrated that four DNA G4C2 repeats can fold into two structurally distinct G4 conformations: a parallel and an antiparallel topology.

Sheep milk has gained increasing attention for its compositional similarity to human milk and its abundance of bioactive compounds with nutritional and therapeutic potential. It is rich in proteins, essential fatty acids, vitamins, minerals, immunoglobulins, and hormones, as well as peptides and oligosaccharides with antiviral, antibacterial, anti-inflammatory, and immune-modulatory effects. Despite these benefits, the literature remains fragmented, with limited integration of data on the mechanisms by which these components influence health outcomes, and few comprehensive comparisons with other mammalian milks.

Although antimicrobial peptides possess potent antimicrobial activities, the high cost of production, based on amino acid length, has limited their therapeutic and cosmeceutical applications. This study aimed to produce and characterize de novo designed antimicrobial peptides derived from WSKK11 and WSRR11 for efficacy against acne-causing bacteria. Ten designed peptides were evaluated for antimicrobial, hemolytic, and cytotoxic activities, as well as, secondary structures by FTIR and modes of action.
