Abstract IL-17 producing CD4 T cells have recently been shown to play an important role in mucosal immunity in HIV infection. But its role in peripheral immunity and the molecular mechanism underlying its regulation during HIV-1 infection are ill defined. In this study, we report a significant negative correlation between IL-17 production in peripheral blood and HIV-1 plasma viral load (pVL). On further investigation, we observe a marked reduction in retinoid-related orphan nuclear receptor (RORγt; Th17 lineage specific transcription factor) binding at IL-17 promoter in HIV patients with high viremia (pVL>10,000 copies/mL) in contrast to relatively low viremic patients which indicate the magnitude of viral copy number on RORγt binding at IL-17 promoter. Additionally, our study highlights that FoxP3 influences IL-17 production by binding to and acting together with RORγt, consequently inhibiting RORγt binding to IL-17 promoter with growing viremia in HIV infection. Collectively, our data suggest that FoxP3 interacts with RORγt transcription factor in a viral load-dependent fashion and brings about negative impact on IL-17 production in HIV-1 infection.

Pemphigus vulgaris (PV) is an autoimmune blistering disorder, which is caused by the loss of desmosomal cell-cell adhesion, initiated by the binding of IgG antibodies against the desmosomal components desmoglein (Dsg)1 and Dsg3. Dsg3-reactive CD4 T helper (Th) cells, in particular follicular Th (Tfh) cells, play a central role in autoantibody production by Dsg3-specific B cells. In this study, we challenged the concept that distinct Dsg3-reactive CD4 T cell subsets are critical in PV pathogenesis utilizing phenotypical and functional state-of-the-art ex vivo assays.

Oral vaccination offers a promising strategy for controlling Helicobacter pylori infection, particularly in the face of rising antibiotic resistance and reinfection rates. In this study, we developed a chitosan nanoparticle-mediated oral DNA vaccine encoding the urease B subunit of H. pylori.

Background: Rhodococcus equi causes pneumonia in young foals, but disease susceptibility and severity vary. Cortisol and vitamin D modulate immune responses and cytokine production during bacterial infection, and altered concentrations are associated with sepsis in neonatal foals. We hypothesised an age and disease effect on circulating steroid hormone concentrations in foals, and that differences in cytokines and steroid hormone concentrations would predict disease severity in pneumonic foals.

Background: Bullous Pemphigoid (BP) is caused by a predominantly Th2-mediated attack on the basement membrane by the production of anti-BP180 and anti-BP230 antibodies. Malignant tumors can exacerbate immune disorders through a variety of potential pathways, including pro-inflammatory responses in the tumor microenvironment, cross-immune responses induced by tumor-associated antigens, and the lifting of immunosuppressive states and activation of underlying autoimmune responses after surgery. Alopecia Areata (AA) is an autoimmune disease caused by T-lymphocyte-mediated destruction of the immune privilege of the hair follicle, specifically involving the immune axes of Th1, Th2 and Th17.

Introduction: Retinoic acid receptor-related orphan receptor gamma t (RORγt) is a crucial transcription factor regulating Th17 cells, which secrete the cytokine IL-17. RORγt inhibitors are regarded as a therapeutic modality in a wide range of autoimmunity including psoriasis.
Objectives: The objective of the study is to investigate novel RORγt inhibitors from natural products (NPs), combining machine learning (ML)-based virtual screening, chemotaxonomic analysis, molecular docking, and molecular dynamics simulations, and biological validation.
