Article Abstract

Reports of oxacillin-susceptible mecA-positive Staphylococcus aureus strains are on the rise. Because of their susceptibility to oxacillin and cefoxitin, it is very difficult to detect them by using routine phenotypic methods. We describe two such isolates that were detected by chromogenic medium and confirmed by characterization of the mecA gene element.

In the present study, we used phenotypic and molecular methods to determine susceptibility to oxacillin in coagulase-negative staphylococci (CoNS) and estimate the prevalence of strains with low-level resistance to oxacillin, A-positive oxacillin-susceptible methicillin-resistant (OS-MRCoNS), and borderline oxacillin-resistant (BORCoNS). One hundred one CoNS strains were screened for oxacillin and cefoxitin susceptibility using phenotypic (disk diffusion, agar dilution, latex agglutination, and chromagar) and molecular (detection of A, B, and C) methods. Staphylococcal cassette chromosome (SCC) typing was performed.

Article Synopsis
  • Understanding the epidemiology of -positive strains is essential for developing treatment strategies and addressing public health risks, focusing on both oxacillin-resistant and susceptible strains in dogs.
  • A study analyzed 87 -positive isolates from dogs with pyoderma in Thailand, revealing that 37.9% were oxacillin-resistant (OR-MRSP), with a majority showing multidrug resistance.
  • Genetic analysis demonstrated that while OR-MRSP strains had consistent antibiogram and SCC types, oxacillin-susceptible strains (OS-MRSP) showed varied patterns, indicating complex colonization dynamics among infected dogs.

The aim of this study is to describe the phenotypic and genetic properties of oxacillin-susceptible methicillin-resistant Staphylococcus aureus (OS-MRSA) isolates and their beta-lactam resistant derivatives obtained after selection with oxacillin. A collection of hospital- (HA-) and community-acquired (CA-) MRSA was screened for oxacillin susceptibility. Antibiotic susceptibility testing, population analysis profile (PAP), mecA expression analysis, and whole genome sequencing (WGS) were performed for 60 mecA-positive OS-MRSA isolates.
