Reprofiling of existing drugs to treat conditions not originally targeted is an attractive means of addressing the problem of a decreasing stream of approved drugs. To determine if 3D shape similarity can be used to rationalize an otherwise serendipitous process, we employed 3D shape-based virtual screening to reprofile existing FDA-approved drugs. The study was conducted in two phases. First, multiple histamine H(1) receptor antagonists were identified to be used as query molecules, and these were compared to a database of approved drugs. Second, the hits were ranked according to 3D similarity and the top drugs evaluated in a cell-based assay. The virtual screening methodology proved highly successful, as 13 of 23 top drugs tested selectively inhibited histamine-induced calcium release with the best being chlorprothixene (IC(50) 1 nM). Finally, we confirmed that the drugs identified using the cell-based assay were all acting at the receptor level by conducting a radioligand-binding assay using rat membrane.

Amidst the global shift toward cannabis legalization, this study examines medical cannabis (MC) sales as an indicator of economic activity and innovation. It explores associations between MC sales, and variables including tobacco use, alcohol consumption, amphetamine, cocaine and cannabis prevalence, and gross domestic product (GDP), using a fixed effects (FE) panel regression model. It also evaluates associations between cannabis legalization and MC sales over time using a dynamic Difference-in-Differences (DiD) approach with multiple time periods.

Nuclear receptors (NRs) are a superfamily of ligand-activated transcription factors that regulate gene expression in response to metabolic, hormonal, and environmental signals. These receptors play a critical role in metabolic homeostasis, inflammation, immune function, and disease pathogenesis, positioning them as key therapeutic targets. This review explores the mechanistic roles of NRs such as PPARs, FXR, LXR, and thyroid hormone receptors (THRs) in regulating lipid and glucose metabolism, energy expenditure, cardiovascular health, and neurodegeneration.

Background: Alcohol use disorder (AUD) is conceptualized as a dimensional phenomenon in the DSM-5, but electronic health records (EHRs) rely on binary AUD definitions according to the ICD-10. The present study classifies AUD severity levels using EHR data and tests whether increasing AUD severity levels are linked with increased comorbidity.
Methods: Billing data from two German statutory health insurance companies in Hamburg included n = 21,954 adults diagnosed with alcohol-specific conditions between 2017 and 2021.

Background: Organotypic long-term cultivation of vascularized retina explants is a major challenge for application in drug development, drug screening, diagnostics and future personalized medicine. With this background, an assay and protocol for organotypic culture of vascularized retina explants in vitro with optimum tissue integrity preservation is developed and demonstrated.
Methods: Morphological, histologic and biochemical integrity as well as viability of vascularized retina explants are compared as function of cultivation time for differently structured nanotube scaffolds.

An Ag-functionalized structural color hydrogel (Ag-SCH) sensor is constructed for colorimetric detection of glutathione (GSH). The hydrogel is prepared by using the coordination of Ag and 1-vinylimidazole (1-VI) as cross-linking network. GSH acts as a competitive ligand to break the coordination between Ag and 1-VI, leading to the expansion and structural color change of the hydrogel.
