Small RNAs and a diverse array of protein partners control gene expression in eukaryotes through a variety of mechanisms. By combining siRNA affinity chromatography and mass spectrometry, we have identified the double-stranded RNA-binding domain protein Blanks to be an siRNA- and dsRNA-binding protein from Drosophila S2 cells. We find that Blanks is a nuclear factor that contributes to the efficiency of RNAi. Biochemical fractionation of a Blanks-containing complex shows that the Blanks complex is unlike previously described RNA-induced silencing complexes and associates with the DEAD-box helicase RM62, a protein previously implicated in RNA silencing. In flies, Blanks is highly expressed in testes tissues and is necessary for postmeiotic spermiogenesis, but loss of Blanks is not accompanied by detectable transposon derepression. Instead, genes related to innate immunity pathways are up-regulated in blanks mutant testes. These results reveal Blanks to be a unique component of a nuclear siRNA/dsRNA-binding complex that contributes to essential RNA silencing-related pathways in the male germ line.

Mast cell tumors (MCTs) are the most common skin neoplasms in dogs and exhibit highly variable biological behavior. Metastasis primarily affects the lymph nodes, though less frequently, MCTs can infiltrate the spleen, liver, peripheral blood, and bone marrow. Flow cytometry of fine needle aspirate samples represents a non-invasive diagnostic procedure that has shown promise for detecting and quantifying mast cells in primary tumors and lymph nodes.

Plant-derived extracellular vesicles have presented great potential in drug and/or nutrition delivery, but it is still unclear whether the variety affects the physicochemical properties of plant derived extracellular vesicles. In this work, the extracellular vesicles from various oranges were first characterized, including navel orange juice (NOJ), green orange juice (GOJ), bingtang orange juice (BTOJ) and blood orange juice (BOJ). The results exhibited obvious distinctions of extracellular vesicles among different oranges, such as vesicle concentration, surface potential, lipid composition, protein content and so on.

Enoxaparin sodium (ES), a low molecular weight heparin derivative, has recently been recognized for its diverse biological activities. In particular, the ability of heparin to modulate inflammation has been utilized to enhance the biocompatibility of bone implant materials. In this study, we utilized poly (methyl methacrylate) (PMMA), a drug loading bone implant material, as a matrix and combined this with enoxaparin sodium (ES) to create enoxaparin sodium PMMA cement (ES-PMMA) to investigate the regulatory effects of ES on inflammatory responses in bone tissue from an animal model.
