The events resulting in compaction have an important influence on the processes related to blastocyst formation. To analyse the quality of the embryos obtained by somatic cell nuclear transfer (SCNT) in aspects different from previous studies, not only the number of blastomeres of cloned embryos during the initiation of compaction, but also the distribution of microvilli in cloned, normal, parthenogenetic, and tetraploid embryos before and after compaction was preliminarily investigated in mouse. Our results showed that during compaction the number of blastomeres in SCNT embryos was fewer than that in intracytoplasmic sperm injection (ICSI) embryos and, before compaction, there was a uniform distribution of microvilli over the blastomere surface, but microvilli became restricted to an apical region after compaction in the four types of embryos. We also reported here that the time course of compaction in SCNT embryos was about 3 h delayed compared with that in ICSI embryos, while there was no significant difference between SCNT and ICSI embryos when developed to the 4-cell stage. We concluded that: (i) the cleavage of blastomeres in cloned embryos was slow at least before compaction; (ii) the distribution of microvilli in cloned, normal, parthenogenetic, and tetraploid embryos was coherent before and after compaction; and (iii) the initiation of compaction in SCNT embryos was delayed compared with that of ICSI embryos.

High-resolution nano-focus X-ray fluorescence microscopy using hard X-rays at the European Synchrotron Radiation Facility (ESRF) IDB16 beamline detected endogenous barium, bromine, calcium, chlorine, copper, iron, manganese, potassium, phosphorus, rubidium, sulphur, selenium, strontium and zinc, at tissue, cellular and subcellular level in the outer retinal complex of light adapted, 3-week-old, male C57BL6 mice. Fresh snap-frozen (20 μm) cryosections dried at room temperature were scanned at 1 μm, 300 nm and 50 nm spatial resolution by incident X-ray photons from the synchrotron beam. Analysis of 2D maps and 3D surface plots by PyMCA and ImageJ revealed elevated zinc concentrations in the choriocapillaris (CC) (mean 45, range 28-77 ppm), retinal pigment epithelium (RPE) layer (mean 47, range 20-76 ppm), photoreceptor inner segments (RIS) ellipsoid zone, outer limiting membrane (OLM) (mean 32, range < 1-44 ppm) and outer nuclear layer (ONL) in between photoreceptor cell bodies.

Gustation or taste in elasmobranch fishes (sharks, skates, and rays) is an important sensory modality that dictates the palatability and ultimately the final decision regarding the ingestion of food. However, the surface morphology, size, abundance, and distribution of taste papillae in this group of apex predators has received little attention. This comparative study uses scanning electron microscopy, histology, and quantitative topographic analyses to assess the surface ultrastructure and density of taste papillae within the oropharyngeal cavity of six batoid species from three families and five selachian species from three families, all from a range of habitats and with a variety of diets.

The mammalian auditory system relies on the precise architecture of the hair cell stereociliary bundle for effective sound transduction. Each bundle consists of approximately 100 actin-filled stereocilia arranged in a three-row staircase pattern, forming a linear shape in inner hair cells (IHCs) and a V-shape in outer hair cells (OHCs), the latter geometry being a hallmark of the mammalian cochlea. While the initial development from uniformly distributed microvilli into stereociliary bundles is guided by lateral migration of the kinocilium, the mechanisms that establish the characteristic bundle architecture and its functional significance remain unclear.
