Article Abstract

C-Terminal carboxylic acid containing inhibitors of the NS3 protease are reported. A novel series of linear tripeptide inhibitors that are very potent and selective against the NS3 protease are described. A substantial contribution to the potency of these linear inhibitors arises from the introduction of a C8 substituent on the B-ring of the quinoline moiety found on the P2 of these inhibitors. The introduction of a C8 methyl group results not only in a modest increase in the cell-based potency of these inhibitors but more importantly in a much better pharmacokinetic profile in rats as well. Exploration of C8-substitutions led to the identification of the bromo derivative as the best group at this position, resulting in a significant increase in the cell-based potency of this class of inhibitors. Structure-activity studies on the C8-bromo derivatives ultimately led to the discovery of clinical candidate 29 (BI 201335), a very potent and selective inhibitor of genotype1 NS3 protease with a promising PK profile in rats.

Herein, we report a visible light-mediated, metal- and photocatalyst-free, green and sustainable synthesis of biologically relevant functionalized benzopyrans by a one-pot, pseudo-multicomponent reaction at room temperature. The notable features of the present protocol include a green and renewable energy source, high yields, easy isolation of the products without performing column chromatography, metal- and photocatalyst-free conditions, room-temperature synthesis, broad substrate scope, environmental friendliness and gram-scale synthesis of the products. In the present study, the benzopyrans are also subjected to molecular docking, which revealed that some benzopyrans are promising potential inhibitors of key nsPs in both Chikungunya and Zika viruses.

We present the discovery of pyrazole-3-carboxylic acid derivatives as novel dengue virus (DENV) NS2B-NS3 protease inhibitors. The discovery was triggered by omission of the phenylglycine scaffold of previous lead structures. We established a robust, regioselective synthetic route toward pyrazole-3-carboxylic acid derivatives.

Hepatitis C virus NS3 helicase contributes to (-) strand RNA synthesis.

Many positive strand RNA viruses encode helicases, but their distinct functions in viral replication cycles is poorly understood. Here, we identify a mutation in the helicase domain of HCV non-structural protein 3 (NS3h), D1467G, which specifically affects (-) strand synthesis, phenocopying mutations in the 3' untranslated region of the genome. D1467G does not impair helicase activity in vitro or the binding of NS3h to critical cis-acting RNA elements, but reduces the interaction of NS3h and NS5B polymerase, potentially contributing to defective (-) strand synthesis.

This study evaluated the safety, tolerability, and pharmacokinetics (PK) of FDV after multiple rising doses in healthy male volunteers and subjects with Gilbert syndrome (GS). In the era of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the application of zinc has increased worldwide. It has the potential to increase the body's antioxidant status and provide better immune makeup. Currently, zinc is indiscriminately used as a therapeutic agent, and this creates an unwanted antagonism with the other interacting micronutrients that are present in the gut and causes secondary deficiencies of other critical micronutrients, which often leads to various complications.
