Article Abstract

Objective: To study whether Notch signaling, which regulates cell fate decisions and vessel morphogenesis, controls lymphatic development.

Methods And Results: In zebrafish embryos, sprouts from the axial vein have lymphangiogenic potential because they give rise to the first lymphatics. Knockdown of delta-like-4 (Dll4) or its receptors Notch-1b or Notch-6 in zebrafish impaired lymphangiogenesis. Dll4/Notch silencing reduced the number of sprouts producing the string of parchordal lymphangioblasts; instead, sprouts connecting to the intersomitic vessels were formed. At a later phase, Notch silencing impaired navigation of lymphatic intersomitic vessels along their arterial templates.

Conclusions: These studies imply critical roles for Notch signaling in the formation and wiring of the lymphatic network.

The nucleolus, traditionally known for its role in ribosome biogenesis, is now recognized for its broader functions, including cellular stress adaptation and its involvement in various pathological processes, such as ribosomal alterations, viral infections, autoimmune disorders, and age-related diseases. Disruptions in nucleolar function can impair protein synthesis, cellular homeostasis, and immune responses, leading to multisystem disorders and increased susceptibility to neoplasms. This review classifies nucleolus-associated diseases into seven categories: deficiencies in protein synthesis, ribosomal and non-ribosomal alterations, cancer and nucleolar alterations, diseases related to aging and cellular stress, autoimmune diseases, and viral diseases.

Bow-tie architecture (BTA) is widely observed in biological neural systems, yet the underlying mechanism driving its spontaneous emergence remains unclear. In this study, we identify a novel formation mechanism by training multi-layer neural networks under biologically inspired non-negative connectivity constraints across diverse classification tasks. We show that non-negative weights reshape network dynamics by amplifying back-propagated error signals and suppressing hidden-layer activity, leading to the self-organization of BTA without pre-defined architecture.
