Article Abstract

Background: MMP-2 and TIMP-2 play important roles in the pathogenesis of arrhythmogenic atrial remodeling, and may contribute to the development and persistence of atrial fibrillation (AF). Functional polymorphisms in the promoter of MMP-2 and TIMP-2 gene may modulate an individual's susceptibility to AF.

Methods: A total of 881 hypertensive heart disease patients from Chinese Han population (128 with and 753 without AF) were recruited in this study. The genotypes of the MMP2-1306C>T and -735C>T polymorphisms and TIMP-2 -418G>C polymorphisms were determined using PCR based method. The plasma concentration of TIMP-2 was measured by enzyme-linked immunosorbent assay in a subgroup with 81 patients.

Results: Both genotype distribution and allele frequency of the TIMP-2 -418G>C polymorphism were significantly different between the AF and control group (P=0.005 and P=0.001, respectively). The C allele carriers (GC+CC) had a significantly increased risk of AF compared with the GG homozygotes (odds ratio,1.77, 95% CI 1.21-2.92, P=0.009) in a logistic regression model after adjustment for age, left atrial dimension, left ventricular mass index, and antihypertensive drugs. The C allele carriers also had reduced levels of plasma TIMP-2 levels compared with GG homozygotes in both AF patients and control subjects. No relationship was found in this cohort between the presence of the MMP-2 -1306C>T and -735C>T polymorphism and AF.

Conclusions: The TIMP-2 -418G>C polymorphism is significantly associated with an increased susceptibility to AF in Chinese Han patients with hypertensive heart disease. The -418C allele, which is associated with a decreased expression of TIMP-2, might be a genetic risk for the development of AF in this cohort.

This study examined the pH-dependent (3, 5, and 7) regulation of matrix metalloproteinase (MMP) activity by cathepsin K (catK) and glycosaminoglycans (GAGs) using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), fluorescence assays, and human dentin slice experiments. The direct effects of catK were evaluated in the catK-active, catK-deficient, and odanacatib (ODN)-inhibited groups, whereas indirect GAG/ tissue inhibitor of metalloproteinase (TIMP)-mediated regulation was assessed in the catK-active, ODN-inhibited, and chondroitin sulfate (CS)-treated groups through dimethylmethylene blue (DMMB) assays, in situ zymography, and immunofluorescence staining. CatK directly activated MMP-2 (62 kDa) and MMP-9 (82 kDa) at all pH values, with no activation observed in the ODN-inhibited or catK-deficient groups.

Purpose: This study aimed to investigate the expression of tissue inhibitor of matrix metalloproteinase-2 (TIMP-2) in postoperative patients with hepatocellular carcinoma (HCC), its relationship with prognosis, and to build a nomogram prediction model for overall survival (OS) and disease-free survival (DFS) based on TIMP-2 expression.

Patients And Methods: Expression profile data from HCC-related datasets were obtained from the Gene Expression Omnibus to analyze the correlation between TIMP-2 and HCC survival and prognosis, and its relationship with the HCC tumor immune microenvironment. Overall, 118 patients who underwent radical surgery for HCC were included retrospectively.

Background/aim: Acute lymphoblastic leukemia (ALL) is the most common pediatric hematologic malignancy, particularly affecting children aged 2~5 years. Tissue inhibitor of metalloproteinase-2 (TIMP-2), a key regulator of MMP-2 activity, has been implicated in several cancers, yet its genetic role in childhood ALL remains unexplored.

Materials And Methods: This study investigated four polymorphic genotypes, rs8179090, rs4789936, rs2009196, and rs7342880, in 266 Taiwanese children with ALL and 266 matched controls using polymerase chain reaction-restriction fragment length polymorphism methodology.

Background: The pathogenesis of aortic aneurysm (AA) is characterized by chronic inflammation of the aortic wall, the associated accumulation of macrophages, and degradation of the extracellular matrix, including elastin. Colchicine (COL) has long been known for its anti-inflammatory effects, so in this study we investigated its effects on AA.

Methods And Results: In vitro, tumor necrosis factor (TNF)-α-stimulated macrophages and vascular smooth muscle cells (VSMCs) were treated with and without COL for 24 h.

Objective: To determine the mechanism of electro-acupuncture (EA) effect by the wingless-related integration site (Wnt)/β-catenin pathway in the guinea pig myopia model.

Methods: Following myopia induction and EA, guinea pigs were treated with biometry to evaluate refraction and axial length. Hematoxylin and eosin (HE) staining was used to observe that the retina, choroid, and sclera had abnormal morphology.
