N-Acylethanolamines, including N-palmitoyl-ethanolamine (PEA), are hydrolyzed to the corresponding fatty acids and ethanolamine by fatty acid amide hydrolase (FAAH). Recently, N-acylethanolamine-hydrolyzing acid amidase (NAAA) was identified as being able to specifically hydrolyze PEA. In order to find selective and effective inhibitors of this enzyme, we synthesized and screened several amides, retroamides, esters, retroesters and carbamates of palmitic acid (1-21) and esters with C15 and C17 alkyl chains (22-27). Cyclopentylhexadecanoate (13) exhibited the highest inhibitory activity on NAAA (IC(50)=10.0 microM), without inhibiting FAAH up to 50 microM. Compound 13 may become a useful template to design new NAAA inhibitors.

Background: As one of the most destructive and invasive pests for various plants in China, Spodoptera frugiperda (Lepidoptera: Noctuidae) poses an enormous threat to food security and results in serious economic losses for harvesting and consumption of agricultural vegetables. To this end, indoxacarb has shown great promise as an effective insecticide against Spodoptera frugiperda. It is metabolized by insect esterases or amidases into the N-decarbomethoxy metabolite (DCJW), which is a key metabolite responsible for the insecticidal activity of indoxacarb.

Cross-linked enzyme aggregates (CLEAs) offer a promising strategy for enzyme immobilization, enhancing reusability, high enzyme loading capacity, carrier-free nature, enhanced thermal or operational stability, and ease of preparation and suitability for industrial-scale applications. This study presents the development and optimization of CLEAs of crude amidase from Bacillus smithii IIIMB2907 for efficient biocatalytic synthesis of pharmaceutically important hydroxamic acids from amides. By optimizing key preparation parameters, a robust immobilized biocatalyst was established that retains high activity and stability during repeated use.

The emergence of beta-lactamase producing multidrug-resistant (MDR) gram-negative bacteria presents a significant challenge to effective treatment of infections. This study focuses on the isolation, amplification, and molecular characterization of β-lactamase genes from clinical strains of Escherichia coli and Klebsiella pneumoniae. Seven new partial gene sequences, including novel variants of blaOXA and blaNDM, were identified after screening 108 clinical samples and submitted to NCBI GenBank.
