We report a catalog of the mouse embryonic pituitary gland transcriptome consisting of five cDNA libraries including wild type tissue from E12.5 and E14.5, Prop1(df/df) mutant at E14.5, and two cDNA subtractions: E14.5 WT-E14.5 Prop1(df/df) and E14.5 WT-E12.5 WT. DNA sequence information is assembled into a searchable database with gene ontology terms representing 12,009 expressed genes. We validated coverage of the libraries by detecting most known homeobox gene transcription factor cDNAs. A total of 45 homeobox genes were detected as part of the pituitary transcriptome, representing most expected ones, which validated library coverage, and many novel ones, underscoring the utility of this resource as a discovery tool. We took a similar approach for signaling-pathway members with novel pituitary expression and found 157 genes related to the BMP, FGF, WNT, SHH and NOTCH pathways. These genes are exciting candidates for regulators of pituitary development and function.

Isolated ectopic secretion of corticotropin-releasing hormone (CRH) is an exceedingly rare cause of Cushing's syndrome (CS), accounting for fewer than 1% of cases. Ectopic CS is an uncommon but potentially life-threatening condition that often necessitates urgent diagnostic evaluation and treatment. Hormonal testing may suggest a pituitary origin, complicating the diagnostic process.

Inborn errors of immunity (IEI) presenting with immunodeficiency and autoimmunity can illuminate pathways essential for immunocompetence and self-tolerance. We recently characterized a new IEI named MAGIS ("idline malformations of the brain, nterior pituitary gland dysfunction, rowth retardation, mmunodysregulation/immunodeficiency, and keletal defects") caused by heterozygous germline activating mutations in (encoding the heterotrimeric G-protein, G). This disorder demonstrates the central role of G regulation of chemotaxis in humans and a novel pathway by which G-proteins regulate T-cell activation.

The low dose dexamethasone stimulation test (LDDST) is routinely used in canine medicine but in the few cases in guinea pigs diagnosed with hypercortisolism, the adrenocorticotropic hormone (ACTH) stimulation test was used. The objective of the authors was to conduct a pilot study and find out if the standard test used in dogs can be used in this species.A 4-year-old intact female hairless guinea pig showed bilaterally enlarged adrenal glands and high cortisol levels.
