The recently published X-ray structures of the beta(2)-adrenergic receptor are the first examples of ligand-mediated GPCR crystal structures. We have previously performed computational studies that examine the potential viability of these structures for use in drug design, exploiting known ligand activities. Our previous study and a newly reported beta(2)/Timolol X-ray complex provide validation of the computational approaches. In the present work, we use the X-ray structures to extract, via in silico high-throughput docking, compounds from proprietary and commercial databases and demonstrate the successful identification of active compounds by radioligand binding.

Background: Studies on the interaction of cancer cells with other cells (fibroblasts, endothelial cells, and immune cells) of the tumor microenvironment (TME) have led to the development of many novel targeted therapies. More recently, the notion that neuronal cells of the TME could impact various processes supporting cancer progression has gained momentum. Tumor-associated neurons release neurotransmitters into the TME that, in turn, bind to specific receptors on different target cells, supporting cancer progression.

Objective: Building upon prior research, our investigation focused on examining changes in gene expression of Connexins 37 and 43 (Cx) influenced by β2-adrenergic agents in cumulus cell cultures from women with polycystic ovarian syndrome (PCOS) and poor ovarian response (POR), all of whom were candidates for in vitro fertilization (IVF).
Materials And Methods: This experimental study was conducted between April 2021 and November 2023, involving three groups: a control group (donated eggs) and two study groups (POR and PCO). All three groups received ovulation stimulation drugs.

The sympathetic nervous system modulates bone mass in part by inhibiting bone formation through the osteoblastic β2-adrenergic receptor (β2AR), but understanding of the bone anabolic processes hindered by β2AR signaling is incomplete. Canonical Wnt signaling is anabolic in bone, promoting osteoblastic bone formation when the multifunctional β-catenin protein is unphosphorylated at N-terminal residues. In the present study, osteoblastic consequences of Wnt-independent β-catenin C-terminal phosphorylation downstream of β2AR activation were investigated.
