Article Abstract

The regulation of the gene encoding the extracellular exo-beta-1,3-glucanase (tag83) produced by the mycoparasite Trichoderma asperellum was studied. Enzyme activity was detected in all carbon sources, but the highest levels were found when starch and purified cell walls from Rhizoctonia solani were used. These results are supported by the appearance of one strong band with enzyme activity in non-denaturing PAGE. Experiments using RT-PCR showed that exo-beta-1,3-glucanase induction in T. asperellum occurred at the transcriptional level. We used RT-PCR and real-time PCR analysis to examine the expression of tag83 gene during in vivo assay of T. asperellum against R. solani. We showed that the expression of tag83 is significantly increased by the presence of R. solani.

In order to investigate the effects of Acer truncatum endophytic fungi Chaetomium globosum and Trichoderma asperellum on root configuration and growth of cucumber seedlings, "Xinjin No. 4" was used as experimental material and pot experiment was conducted. The experiment was set up with four treatments: a blank control (CK), a treatment with Chaetomium globosum at 100% concentration (CG), a treatment with Trichoderma asperellum at 100% concentration (TA), and a mixed strain treatment combining 50% Chaetomium globosum and 50% Trichoderma asperellum administered via root irrigation (GA).

This study, conducted over two consecutive seasons in an organic-certified Mediterranean tomato greenhouse, aimed to assess the dynamics of soil bacterial composition at pre-planting phase following the incorporation of fresh sheep manure and subsequent solarisation (biosolarisation), as well as at the end of the crop cycle.
