Acetylcholinesterase activity in defined brain regions was determined using biochemical and histochemical methods 30 min after treating rats with sarin, soman or VX (0.5 x LD(50)). Enzyme inhibition was high in the pontomedullar area and frontal cortex, but was low in the basal ganglia. Histochemical and biochemical results correlated well. Determination of the activity in defined brain structures was a more sensitive parameter than determination in whole brain homogenate where the activity was a "mean" of the activities in different structures. The pontomedullar area controls respiration, so that the special sensitivity of acetylcholinesterase to inhibition by nerve agents in this area is important for understanding the mechanism of death caused by nerve agents. Thus, acetylcholinesterase activity is the main parameter investigated in studies searching for target sites following nerve agent poisoning.

This study aimed to develop an efficient green ultrasound-assisted extraction (UAE) method for naringin (Nar) from Exocarpium Citri Grandis (ECG) using a glycerol-based ternary natural deep eutectic solvent (NADES) and explore its biofunctional relevance. After screening and single-factor optimization, the optimal NADES was identified as glycerol:malic acid:propanedioic acid (1:1:2 M ratio, 30 % water content). Extraction conditions (liquid-solid ratio, temperature, time) were optimized via response surface methodology (RSM) and an artificial neural network-genetic algorithm (ANN-GA), with ANN-GA demonstrating superior predictive capability.

A strategically engineered, eco-conscious synthetic platform was developed to access a novel library of eighteen polyfunctionalized pyridine-based heterocycles through high-efficiency multicomponent and annulation strategies, using 2-amino-4-(4-chlorophenyl)-6-(p-tolyl)nicotinonitrile (M) as a privileged core. Structural diversity was maximized by integrating potent pharmacophores, including pyrido[2,3-d]pyrimidines, naphthyridines, triazines, and fused pyrrolo/tetrazolo motifs, via both conventional and accelerated (microwave/ultrasound-assisted) routes, affording excellent yields with high structural fidelity as confirmed by IR, H/C NMR, and mass spectrometry. Biological evaluation revealed that all synthesized compounds had excellent larvicidal efficacy against Culex pipiens larvae, especially 15 and 9, emerging as lead candidates that exhibited exceptional LC₅₀ values of 0.

Introduction: Arsenic, a metalloid, is well associated as a risk factor for the development and progression of neurodegenerative diseases, including Alzheimer's Disease (AD), which is characterized by impairment in cognition. However, specific effects of arsenic on Acetylcholinesterase (AChE) activity and inflammatory markers in different brain regions, as well as its impact on behaviour, are not yet fully understood.
Methods: Arsenic was administered (20 mg/kg by gavage for 4 weeks) to male and female mice, and its effects on behaviour were assessed by using the object recognition memory test and lightdark box test.
