Article Abstract

Purpose: Extensive remodeling of the lamina cribrosa extracellular matrix occurs in primary open angle glaucoma. The transforming growth factor-beta (TGF-beta) and matrix metalloproteinase (MMP) protein families are implicated in this process. The authors investigated (a). the effect of cyclical mechanical stretch on TGF-beta1 mRNA synthesis, TGF-beta1 protein secretion, MMP-2 protein activity and (b). the effect of exogenous TGF-beta1 on MMP-2 protein activity in human lamina cribrosa cells in vitro.

Methods: Primary human lamina cribrosa cells grown on flexible and rigid plates were exposed to cyclical stretch (1Hz, 15%) or static conditions for 12 and 24 hours. Cells grown on 100-mm plates were exposed to human TGF-beta1 (10 ng/ml) or vehicle (4 mM HCl/1% BSA) for 24 hours. TGF-beta1 mRNA synthesis in stretched and static cells was measured using real-time polymerase chain reaction. TGF-beta1 protein secretion in stretched and static cell media was measured using enzyme linked immunosorbent assay. Gelatin zymography measured MMP-2 activity in stretched, static, TGF-beta1- treated and vehicle-treated cell media.

Results: Cyclical stretch induced significant increases in TGF-beta1 mRNA synthesis after 12 hours (**P < 0.01) and TGF-beta1 protein secretion after 24 hours (*P < 0.05). Cyclical stretch significantly (*P < 0.05) increased MMP-2 activity in cell media after 24 hours. Exogenous TGF-beta 1 induced a significant (**P < 0.01) increase in cell media MMP-2 activity after 24 hours.

Conclusions: These results suggest that cyclical stretch and TGF-beta1 modulate MMP-2 activity in human lamina cribrosa cells. TGF-beta 1 and MMP-2 release from lamina cribrosa cells may facilitate matrix remodeling of the optic nerve head in primary open angle glaucoma.

Aim: To propose a novel glaucoma surgery for rebalancing translaminar pressure difference.

Methods: Three non-human primates with normal eyes and two with laser-induced glaucoma underwent the novel surgical procedure. Cannulation of the subarachnoid space was performed after completion of routine vitrectomy steps.

Purpose: To investigate the role of the lamina cribrosa (LC) astrocytes in the autoregulatory capacity of optic nerve head (ONH) vessels and to explore the underlying molecular mechanisms.

Methods: The Oxygen-glucose deprivation/reperfusion model (OGD/R) was constructed to examine the changes in cell morphology and protein expression in LC astrocytes. LC astrocytes were co-cultured with vascular smooth muscle cells (VSMCs) to detect the role of LC astrocytes in the autoregulatory function of vessels.

Harran University, Faculty of Medicine, Department of Ophthalmology, Şanlıurfa, Türkiye. Electronic address:

Purpose: To compare lamina cribrosa structural parameters for diabetic retinopathy and evaluate the potential use of the lamina cribrosa curvature index (LCCI) as a biomarker for progression.

Methods: All participants underwent comprehensive ophthalmic examinations and optical coherence tomography (OCT) imaging of the optic nerve head. The following lamina cribrosa parameters were measured from high-resolution B-scan images; LCCI, lamina cribrosa curve depth (LCCD), anterior lamina cribrosa depth (ALCD), and lamina cribrosa thickness (LCT).

Department of Clinical Neurosciences, University of Cambridge, Cambridge, UK; Cambridge Clinical Vision Laboratory, University of Cambridge, Cambridge, UK.

Background: Aberrant myelination of the retinal nerve fibre layer (RNFL) can follow lamina cribrosa damage. Such cases may provide insight into how increased myelin affects the visual evoked potential (VEP), a key outcome in remyelination trials.

Objective: We present the case of a myelinated RNFL in a patient with relapsing-remitting multiple sclerosis (RRMS) with accompanying electrophysiology.

Purpose: To propose a new parameter for evaluating lamina cribrosa (LC) morphology in myopic eyes with optic nerve head (ONH) distortion.

Methods: This study included 169 eyes with normal-tension glaucoma (NTG) and myopic ONH distortion from the Myopic NTG Treatment Study. The LC curvature index (LCCI) and the LC steepness index (LCSI) were assessed in optical coherence tomography B-scan images obtained using an enhanced-depth imaging technique.
