Article Abstract

The migration kinetics of bisphenol-A-diglycidyl-ether (BADGE) from processed and non-processed model cans into vegetable oil was investigated as a function of the process treatment and the temperature of storage. Cans were either not heat-treated at all or were processed at 115 degrees C for 30 min or for 1 h after filling with oil. Each series of experiments comprised 30 samples and was further divided into three groups to be stored at different temperatures (20, 40 and 60 degrees C). Aliquots from the samples were taken at regular intervals for > 1 year. Samples were analysed for BADGE by high-performance liquid chromatography with fluorescence detection. The results showed that temperature processing had the largest effect on migration of BADGE. Storage temperature also significantly influenced migration from non-processed cans, in particular at higher storage temperatures such as 60 degrees C. Some samples were Subjected to 60 degrees C storage after an initial period at 20 degrees C and an effect on migration was also noted, although to a much lesser extent than from processing. The results of migration at higher temperatures were also correlated to the potential degradation of BADGE from oxidation products.

BADGE can easily migrate from containers and result in food contamination; the compound is known as an endocrine-disrupting chemical. We previously reported that maternal exposure to bisphenol A bis (2,3-dihydroxypropyl) ether (BADGE·2HO), which is the most detected BADGE derivative not only in canned foods but also in human specimens, during gestation and lactation, could accelerate neuronal differentiation in the cortex of fetuses and induce anxiety-like behavior in juvenile mice.

Binding mechanism and safety assessment of BADGE from epoxy-coated cans and protein.

Bisphenol A diglycidyl ether (BADGE) from the epoxy coating can migrate to food and react with the protein, but the safety of these adducts formed is unknown, besides, after being consumed in large quantities, BADGE may also combine with other proteins supplemented, posing a potential hazard to human health. The binding mechanism and the possible binding products of BADGE and protein were explored by a series of liquid chromatograph-tandem mass spectrometry (LC-MS) methods. Results showed BADGE can modify a variety of amino acid residues with two modification modes, of which the largest number of modifications were cysteine (Cys) and lysine (Lys).

Bisphenol A diglycidyl ether (BADGE) is widely present in the inner coating of metal food cans, from which it can migrate into food and generate harmful derivatives during storage, such as bisphenol A (2,3-dihydroxypropyl) glycidyl ether, bisphenol A (3-chloro-2-hydroxypropyl) glycidyl ether, and bisphenol A (3-chloro-2-hydroxypropyl) (2,3-dihydroxypropyl) glycidyl ether. Here, a gold-nanoparticle-based immunochromatographic strip assay based on a broad-spectrum polyclonal antibody was developed for the simultaneous detection of BADGE and its derivatives, which could be accomplished within 15 min.
