Budding yeast centromeres are comprised of approximately 125-bp DNA sequences that direct formation of the kinetochore, a specialized chromatin structure that mediates spindle attachment to chromosomes. We report here a novel role for the histone deposition complex chromatin assembly factor I (CAF-I) in building centromeric chromatin. The contribution of CAF-I to kinetochore function overlaps that of the Hir proteins, which have also been implicated in nucleosome formation and heterochromatic gene silencing. cacDelta hirDelta double mutant cells lacking both CAF-I and Hir proteins are delayed in anaphase entry in a spindle assembly checkpoint-dependent manner. Further, cacDelta and hirDelta deletions together cause increased rates of chromosome missegregation, genetic synergies with mutations in kinetochore protein genes, and alterations in centromeric chromatin structure. Finally, CAF-I subunits and Hir1 are enriched at centromeres, indicating that these proteins make a direct contribution to centromeric chromatin structures.

RNA localization and local translation are widespread phenomena that play key roles in a plethora of cellular processes ranging from embryo patterning to general cellular functions. The traditional paradigm assigns localization elements to cis-acting RNA sequences which assemble into complexes that regulate mRNA transport and translation, and the mRNA is generally transported while remaining translationally silent. However, recent evidence has shown that the nascent protein can also play an essential role in RNA localization and can enable polysomes to control their own transport and be delivered where and when they are needed.

Objectives: This study aims to predict the severity of Parkinson's disease (PD) by leveraging a comprehensive dataset integrating cerebrospinal fluid protein and peptide data sourced from UniProt, normalized protein expression metrics, clinical assessments, and gait data. The dataset comprised 248 PD patients monitored longitudinally, with periodic evaluations including 227 proteins, 971 peptides, gait parameters, and Movement Disorder Society-sponsored revision of the Unified Parkinson's Disease Rating Scale (MDS-UPDRS) scores at baseline 0, 6, 12, and 24 months.
Methods: A multifaceted machine learning framework was employed, consisting of random forest, TensorFlow decision forests, and a custom-developed phaseshift ensembling model.
