Article Abstract

Isothiouronium and thiazolidinium salts are sulfur-containing scaffolds commonly found in bioactive molecules. We report an expeditive one-pot, two-step procedure for the rapid synthesis of isothiouronium salts from carbon disulfide under microwave irradiation, allowing their isolation in less than 30 min and in good to excellent yields, without the need for a catalyst. When propargyl bromide is used as an alkylating agent, the corresponding isothiouronium salt undergoes an intramolecular cyclization during silica gel chromatography, affording a thiazolidinium salt. This rearrangement, not observed under the reaction conditions, was investigated via DFT calculations. Computations show that the uncatalyzed isomerization is not feasible but becomes accessible in the presence of silica gel, which acts as a proton shuttle. The rearrangement is shown to comprise two main steps that can take place in any order, i.e., [1,3] hydrogen shift and C-N bond formation. This leads to two alternative mechanisms with similar free energy barriers of ca. 18-19 kcal·mol, in both cases associated with the rate-determining C-N bond formation step.

The universal two-stage synthesis of 1-alkyl-4-methyl- and 1,3,4-trialkylsubstituted semiselenoglycolurils was realized on the basis of a range of semithioglycolurils, which were S-methylated to isothiouronium salts, followed by the interaction of the salts with NaHSe generated in situ from Se and NaBH. The resulting semiselenoglycolurils were tested as antimicrobials and exhibited selective inhibition of filamentous fungi. A broad comparison with previously reported analogs revealed their fungistatic mode of action and highlighted the significant influence of steric hindrance of the selenium atom on antifungal activity.

The focus here is on the structural study of iso-thio-uronium salts and the application of intermolecular distances obtained by solid-state NMR (ssNMR) in determining crystal structures from powder diffraction data. The synthesis of three new tetra-fluoro-borate salts and two bromide salts of iso-thio-uronium compounds is presented first, followed by structural and spectroscopic studies. The tetra-fluoro-borates were further analysed using advanced ssNMR techniques to obtain a set of intermolecular F⋯C, B⋯B, H⋯H and C⋯H distances with an estimation of their precision.

A practical and efficient method for the synthesis of thioglycosides has been developed using readily available isothiouronium salts as masked S-glycosylation reagents. Under optimized conditions, per-acetylated glycoside bromides were robustly converted into thioglycosides in moderate to good yields with exclusive stereoselectivity. By employing an increased dosage of TMG(1,1,3,3-tetramethylguanidine), a one-pot S-glycosylation-deacetylation process was achieved, affording deprotected thioglycosides in high yields.

Thioesters are important in synthesis, materials science, and biology, and their preparation traditionally relies on the use of disagreeable thiols. Here, we report a thiol-free protocol that stitches together widespread carboxylic acids and aryl halides, producing a diverse array of thioesters. Crucial to this strategy is the discovery that tetramethylthiourea can serve as both a sulfur source and, upon direct excitation by purple light, as a strong reductant, suitable for activating aryl halides via single-electron transfer.
