The study of active substances in traditional Chinese medicine(TCM) is the foundation of TCM pharmacology, TCM quality control, and new drug development, and it is also one of the most popular directions in TCM modernization research in recent years. Due to their diverse chemical compositions and complex component-effect relationships, Chinese medicines often require the comprehensive use of multidisciplinary technologies such as chemistry, biology, and information science to reveal their active substances and targets. In this paper, we review the innovative breakthroughs made in the past 30 years in the discovery strategies and technological means for the research of active substances in traditional Chinese medicine from the perspective of technological changes, and focus on the new direction of the research of active substances in traditional Chinese medicine under the paradigm shift of scientific research brought about by artificial intelligence, with the aim of promoting research in related fields to move in the direction of more accurate, efficient, and intelligent, and providing innovative ideas for the research of active substances in traditional Chinese medicine under the new situation.

A- and B-type procyanidins (PCs) are widely known for their health-promoting properties, such as antioxidant activity. The limited availability of reference substances represents a major challenge, resulting in a low number of systematic studies on their health benefits. In our study, the optimised 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-induced oxidation of the B-types B1 and B2 was carried out yielding the corresponding A-types A1 (1) and A2 (2), which have an additional ether bridge between C2--C7, whereas oxidation of B3 and B4 produced a six-membered spirocyclic ring system including a spiro-carbon atom at C8t (3-5).

Bioprotective LAB3 cells that produce bacteriocin-like substances were entrapped in 4% (w/w) sodium alginate matrices, either with or without 10% (w/w) sodium caseinate. The effects of bead formulation-alginate alone or combined with caseinate, with or without the addition of 20% (w/w) MRS broth or M17 broth-on the culturability of LAB3 cells within the beads and their anti activity were assessed over 12 days of storage at 30 °C in closed bottles. Calcium-alginate-caseinate beads supplemented with MRS broth proved most effective in preserving both culturability and anti- activity.

The activation of nucleotide oligomerization domain-like receptor (NLR) family, pyrin domain-containing protein 3 (NLRP3) inflammasome is implicated in the pathogenesis of various inflammatory diseases. The natural product oridonin possesses a novel mechanism for NLRP3 inhibition and a unique binding mode with NLRP3, but its poor anti-inflammatory activity limits further application. After virtual screening of diverse natural product libraries, dehydrocostus lactone (DCL) was considered as a potential NLRP3 inhibitor.
