The precise structural and functional characteristics of input circuits targeting histaminergic neurons remain poorly understood. Here, using a rabies virus retrograde tracing system combined with fluorescence micro-optical sectioning tomography, we construct a 3D monosynaptic long-range input atlas of male mouse histaminergic neurons. We identify that the hypothalamus, thalamus, pallidum, and hippocampus constitute major input sources, exhibiting diverse spatial distribution patterns and neuronal type ratios. Notably, a specific layer distribution pattern and co-projection structures of upstream cortical neurons are well reconstructed at single-cell resolution. As histaminergic system is classically involved in sleep-wake regulation, we demonstrate that the lateral septum (predominantly supplying inhibitory inputs) and the paraventricular nucleus of the thalamus (predominantly supplying excitatory inputs) establish monosynaptic connections, exhibiting distinct functional dynamics and regulatory roles in rapid-eye-movement sleep. Collectively, our study provides a precise long-range input map of mouse histaminergic neurons at mesoscopic scale, laying a solid foundation for future systematic study of histaminergic neural circuits.

Background And Objective: NMDA receptors, predominantly located in the central nervous system and known for their roles in synaptic plasticity and central sensitisation of pain and itch, are also expressed in peripheral sensory neurons. Emerging evidence suggests that peripheral NMDA receptors contribute to pathological pain and potentially itch, identifying them as promising therapeutic targets. The aim of this review is to explore the role of peripheral NMDA receptors in pain and itch and to summarise the effectiveness of topical NMDA antagonists in managing these sensations.

Individuals with Down syndrome exhibit various changes in the human body systems, including alterations in the ocular, neurological, and dermatological systems. Especially, preclinical and clinical studies have determined Down syndrome patients to possess reduced intellectual and cognition abilities, which neurobehavioral effects are associated with altered molecular markers in the brain. For instance, neuroinflammation and increased brain oxidative stress are reported in animals models of Down syndrome, and the reversal of those markers lead to positive effects.

Numerous studies in both humans and rodent models have consistently demonstrated the important role of the anterior cingulate cortex (ACC) in emotion perception, pain processing, memory formation, etc. The ACC can be anatomically subdivided into rostral and caudal subregions, each exhibiting distinct functional profiles.
