Article Abstract

Microphysiological systems (MPS) contain multiple cell types in three-dimension and often incorporate fluidic shear forces. There is interest in MPS for disease and efficacy modeling, safety and disposition studies. Animal cell-based MPS are needed to provide confidence in translation of data from human cell-based MPS. We developed rat and dog quad-culture liver MPS incorporating primary hepatocytes, sinusoidal endothelial, Kupffer and stellate cells. Using cryopreserved primary cells, we established a protocol for co-culturing cells under physiological flow conditions. Cells were evaluated for viability, morphology and function (e.g., albumin production, cytochrome P450 and flavin-containing mono-oxygenase (FMO) activity). Optimized culture conditions maintained high-quality rat and dog liver chips for up to 7 days. Model performance was evaluated with ABT-288, a histamine-3 receptor antagonist that caused elevated serum transaminases in dogs but not rats. This finding was partially attributed to high levels of FMO-mediated N-oxide metabolite produced in the dog. Key findings in our study were 1) dog chips showed much higher FMO-mediated N-oxidation compared to rat, and 2) dog chips exhibited modestly higher sensitivity to ABT-288 toxicity endpoints (albumin, alanine transaminase and lactate dehydrogenase) compared to rat. Species differences in N-oxidation were not observed in rat and dog liver microsomes or 2D hepatocyte monocultures, suggesting that properties of the quad-culture MPS were necessary to model higher FMO activity observed in dogs in vivo. The data suggest that this preclinical species liver chip model provides novel understanding of vitro to in vivo translation of ABT-288 dog liver toxicity.

Our objective was to describe a case of suspected relay barbiturate intoxication of a dog after ingestion of a rat that had been euthanized and frozen and then later burned in an attempted cremation. This case will be compared to previous reports of relay toxicosis. This report describes a dog that was presented to an emergency and critical care hospital because of lethargy and vomiting after ingesting remains of a rat that had been euthanized 2 wk earlier.

Amphenmulin is a novel pleuromutilin derivative with proven excellent antibacterial activity. To investigate its metabolism in animals, ultra-high-performance liquid chromatography coupled with quadrupole/time-of-flight mass spectrometry (UHPLC-Q-TOF-MS/MS) was employed to analyze and identify metabolites in rats and chickens and using human, rat, pig, chicken and beagle dog liver microsomes. We identified 18 metabolites from liver microsomes and 24 and 17 metabolites for rats and chickens, respectively.

The neuroendocrine protein 7B2 plays a crucial role in the maturation and activity regulation of prohormone convertase 2 (PC2). To elucidate the relationship between 7B2 and PC2 expression in endocrine tissues, we generated synthetic peptide antibodies in guinea pigs. The antigenic peptide sequences were selected to correspond to three different positions in the rat amino acid (aa) sequence: The -terminal aa 1-14 is situated immediately following the signal sequence, the middle aa 77-90 contains a part of the proPC2 activation domain, and the -terminal aa 156-168 functions to suppress PC2 activity.
