Article Abstract

The production of E, E-farnesol (FOH) from an indigenous Saccharomyces cerevisiae Bot-S4 was introduced in the present study. A total of twelve (Bot-S1 to S12) different strains of S. cerevisiae were isolated from different samples of baker's yeast, using Yeast Malt agar medium (2.5 g/L yeast extract, 2 g/L malt extract, 8 g/L d-glucose, 1.5 g/L peptone, 20 g/L agar), pH 6.2, containing penicillin 0.02 % (w/v), under stationary culture. The colony exhibiting clear and larger zones of FOH formation (∼1.2 mm, dia.) compared with control (plates without ergosterol) was picked up as a hyper producer (BotS4) and selected for optimizations. Different parameters for cultural conditions and analytical techniques, along with critical phases were investigated. The highest FOH production was observed when 50 mL of YM-medium was incubated at 28 °C, pH 5 for 48 h after inoculating with 6 % (v/v) inoculum of 18 h old. FOH production was increased 2-fold by the addition of 0.05 % (w/v) diocetyl ester of sodium sulpho succinic acid (commonly known as MOT or monoxal OT) as a detergent. Glycerol as an alcoholic agent, peptone as a nitrogen source, and olive oil as a fatty acid source also exhibited an inductive effect on the activity when supplemented solely into the reaction mixture. However, the addition of 11.5 % (w/v) combination of MOT + glycerol+peptone+olive oil at (1:1:1:1) supported the highest extra-cellular and cellular FOH fractions (889.28 and 198.45 mg/L, respectively). Overall, more than an 8.5-fold enhancement in FOH production was accomplished. The maximum FPP synthase activity (92.34 U/mL) was also recorded during the course of enzymatic reaction.
