This work presents a comprehensive study on the prediction of phenytoin solubility at supercritical state using advanced techniques including machine learning analysis. The solubility of small-molecule pharmaceutical was analyzed and calculated to enhance its solubility and bioavailability as well. The models were employed to approximate the solubility at various pressures and temperatures. The dataset comprises temperature (T), pressure (P), and solubility (y) values, along with the corresponding solvent density measurements that were used in the models. Three models, namely Automatic Relevance Determination Regression (ARD), Gaussian process regression (GPR), and Linear Regression (LR) were designed and tuned to build predictive models. The ADABOOST ensemble technique was applied to strengthen the predictive capabilities of the models, while hyperparameter tuning was conducted using the Jellyfish Optimization (JO) algorithm. For phenytoin solubility prediction, the ADA-GPR model demonstrated outstanding accuracy, obtaining an R² of 0.99644. The ADA-LR model also produced competitive results, attaining an R² value of 0.93381, whereas the ADA-ARD model showed robust performance, yielding an R² of 0.95249. In terms of solvent density prediction, the ADA-GPR model once again outperformed the others, with an R² value of 0.9933.

Promiscuity, or selectivity on a spectrum, is an encoded feature in biomolecular anion recognition. To unravel the molecular drivers of promiscuous anion recognition, we have employed a comprehensive approach - spanning experiment and theory - with the Staphylococcus carnosus nitrate regulatory element A (ScNreA) as a model. Thermodynamic analysis reveals that ScNreA complexation with native nitrate and nitrite or non-native iodide is an exothermic process.

As the primary storage protein, highland barley gliadin (HBG) exhibits limitations in the processing of highland barley foods, primarily due to its abundant non-polar amino acids. In this study, HBG was utilized to prepare sugar-HBG complexes with pentose (xylose), hexoses (glucose and galactose), and disaccharides (lactose and maltose) in an aqueous system at a pH of 11 and a temperature of 75 °C. Subsequently, the structural and functional characteristics of these complexes were evaluated.

In modern drug discovery, there is a pressing need for rapid, cost-effective, and accessible methods to evaluate the biological activities of newly synthesized compounds. Traditional kinase assay platforms are often labor-intensive, time-consuming, and require specialized equipment or expertise. To address these limitations, we developed and validated a convenient in vitro kinase assay based on a recombinant biosensor, Picchu-B, constructed using a bacterial expression system.

Except from the myeloid in origin cells, ectopic expression of T-cell receptors (TCRs) was also detected in sperm and the female reproductive tract, which in conjunction with major histocompatibility complex (MHC) molecules suggested their involvement in mate choice. Following-up these observations and considering that MHC/TCR interactions could guide spermatozoa towards ovum, the present study aimed to delineate the presence of TCRs in sperm vis-à-vis cognate recognition and define such expression during spermatogenesis. Immunofluorescence experiments using fertile BALB/c males showed that despite the inbred origin of mice, all combinations of high (HI, >20 % expression) and low (LO, <5 % expression) TCRαβ and TCRγδ expression could be detected in equal distribution rates, followed by an inverse pattern of MHC expression.

The soil in reclaimed shale gas sites is compacted and suffers from issues like poor drainage, drought conditions, and nutrient deficiency, posing challenges for agricultural production. In this study, rare earth tailings were incorporated into biochar at different mass ratios (rare earth tailings: biochar = 1:1, 1:2, 1:3, 1:4). Subsequently, a series of rare earth tailings-doped biochar materials (REE-BC) were prepared by calcination at 700°C.
