Article Abstract

Background: BK polyomavirus-associated nephropathy (BKVAN) is a major cause of graft dysfunction in kidney transplant recipients, and is often triggered by BK virus reactivation due to immunosuppression. This study used GeoMx digital spatial profiling (DSP) to investigate molecular changes during BK virus infection.

Methods: Eight formalin-fixed and paraffin-embedded (FFPE) kidney samples from the following three groups were analyzed: the normal function (n=3), BK polyomavirus viremia (BKV) (n=2), and BKVAN (n=3) groups. Transcriptional changes in epithelial cells, immune cells, and fibroblasts were assessed. Immune microenvironment alterations were analyzed through cellular deconvolution.

Results: Distinct molecular signatures were observed across the infection stages and cell types, of which, epithelial cells showed significant dysregulation and unique involvement in ribosomal protein synthesis. The pathway analysis revealed that the pathways associated with viral infection, allogeneic rejection, and immunity (antigen presentation, chemokines, and inflammation) were significantly enriched during BKVAN progression. The key genes associated with BK polyomavirus (BKPyV) infection (, , , , , , , and ), progression (, , , and ), and BKVAN specificity (, , , and ) were identified. The CD74 major histocompatibility complex (MHC) class II axis may serve as a key immunoregulatory pathway. Cellular deconvolution revealed increased macrophages, dendritic cells (DCs), and CD4/CD8 memory T cells, and reduced natural killer (NK) cells and neutrophils in BKVAN.

Conclusions: DSP highlighted inter- and intra-patient heterogeneity, offering insights into the molecular mechanisms underlying BKVAN and potential targets for therapeutic intervention.

Photoremovable protecting groups (PRPGs) enable precise spatiotemporal control over molecular release and functional activation. Recent advances have introduced wavelength-selective systems for sequential deprotection, broadening applications in drug delivery, material synthesis, and photopolymerization. In parallel, PRPGs play a crucial role in photobase generators (PBGs) and photoacid generators (PAGs), enabling oxygen-tolerant, spatially controlled polymerization and depolymerization through light-induced base and acid release.

Conservation has embraced advances in big data and related digital technologies as key to preventing biodiversity loss, especially in the identification of areas of conservation priority based on spatial data, which we call the big geospatial data turn. This turn has led to the proliferation of useful methods and tools, including global geospatial maps. But these methods may also undermine moves toward rights-based and inclusive conservation approaches that consider plural values and perspectives.

There is limited understanding of the impact of anti-IL5 treatment on nasal polyp tissue biology in chronic rhinosinusitis with nasal polyps (CRSwNP). This study examined nasal polyp tissue cellular proteome and transcriptome responses to anti-IL5 treatment in CRSwNP utilising spatial profiling. GeoMx™ Digital Spatial Profiling (DSP) of 80 proteins and 1,833 mRNA targets in the polyp stroma and the whole transcriptome (18,815 mRNA targets) in polyp epithelia was undertaken on sinonasal biopsies collected from 20 individuals with eosinophilic CRSwNP before and after 16 and 24 weeks of mepolizumab treatment.

Objective: Accurate segmentation of breast lesions, especially small ones, remains challenging in digital mammography due to complex anatomical structures and low-contrast boundaries. This study proposes DVF-YOLO-Seg, a two-stage segmentation framework designed to improve feature extraction and enhance small-lesion detection performance in mammographic images.

Methods: The proposed method integrates an enhanced YOLOv10-based detection module with a segmentation stage based on the Visual Reference Prompt Segment Anything Model (VRP-SAM).
