Article Abstract

The review introduces the challenges and potential solutions in xenotransplantation, focusing on pig-to-human organ transplant. Xenotransplantation, mainly with the use of pig organs, is a promising solution because of the reproductive capacity, size, and physiological resemblance of pigs to humans. However, immunological barriers, especially humoral and cellular immune responses, pose substantial challenges. The humoral immune response, involving antibodies targeting xenoantigens, is a substantial barrier. Anti-α-galactose antibodies, targeting α-Gal epitopes, are crucial in hyperacute rejection and acute humoral xenograft rejection. Genetic modifications, including CRISPR/Cas9 technology, aim to eliminate xenoantigens like α-Gal, potentially overcoming these challenges. This review discusses the use of genetically modified pigs for xenotransplantation, emphasizing the removal of xenoantigens, expression of human complement regulatory proteins, and transgenic expres-sion of human regulatory factors. Recent advancements, such as the world's first porcine-to-human heart transplant, highlight the potential of genetic manipu-lation in overcoming immune rejection barriers.

Host-pathogen interactions involve two critical strategies: resistance, whereby hosts clear invading microbes, and tolerance, whereby hosts carry high pathogen burden asymptomatically. Here, we investigate mechanisms by which Salmonella-superspreader (SSP) hosts maintain an asymptomatic state during chronic infection. We found that regulatory T cells (Tregs) are essential for this disease-tolerant state, limiting intestinal immunopathology and enabling SSP hosts to thrive, while facilitating Salmonella transmission.

Adaptation of intestinal helminths to vertebrates involved the evolution of strategies to attenuate host tissue damage to support parasite reproduction and dissemination of offspring to the environment. Helminths initiate the IL-25-mediated tuft cell-type 2 innate lymphoid cell (ILC2) circuit that enhances barrier protection of the host, although viable parasites can target and limit this pathway. We used IL-25 alone to create small intestinal adaptation, marked by anatomic and immunologic changes that persisted months after induction.

Aquatic environmental pollution could be a direct trigger of infection through cercarial invasion to skin / gills or indirectly as a predisposing factor that damage the physical barriers of targeted fish resulting in high intensities of EMC infections in all fish vital organs. In the current study, a total of 150 African catfish (Clarias gariepinus) were randomly collected from Mariotteya drain all the way through the Egyptian township of Shabramant located at the historical heart of Giza. Catfish samples were collected in mid-summer during the period from June to July 2024.

Gut mucin fucosylation dictates the entry of botulinum toxin complexes.

Botulinum toxins (BoNTs) are the most potent known bacterial toxins. The BoNT complex from B-Okra (large progenitor toxin complex (L-PTC)/B, hyper-oral-toxic) exerts at least 80-fold higher oral toxicity in mice compared with that from serotype A1 (L-PTC/A, non-hyper-oral-toxic). Here, we showed that L-PTC/B was predominantly absorbed through enterocytes, whereas L-PTC/A targeted intestinal microfold cells.
