Article Abstract

Gelatin zymography is a simple semiquantitative technique for assaying gelatinases in various biological samples. It is particularly feasible to measure two gelatin-degrading matrix metalloproteinases, MMP-2 (gelatinase A) and MMP-9 (gelatinase B). The use of SDS-polyacrylamide gels impregnated with gelatin provides a reliable way to detect and quantify the type, amount, and activation level of gelatinases in biological samples (cell lysates, tissue extracts, and biological fluids). This method can be used to investigate the expression profile and activation level of gelatinase in cancer cells or tissues and to determine how these changes are related to cancer progression. However, it is crucial to understand the principle and limitation of gelatin zymography before interpreting the experimental data. Gelatin zymography is a powerful method for assaying gelatinases in cancer research if some precautions are taken.

BackgroundWe explored the potential impact of testosterone treatment in a male rat model of chronic obstructive pulmonary disease (COPD). Our study focused on evaluating the potential decrease in the expression and activation of matrix metalloproteinase-9 (MMP-9) and fibroblast growth factor-23 (FGF-23) induced by COPD.MethodsWistar rats were randomly assigned to one of three groups: control, COPD, or testosterone treatment.

Metastatic melanoma is a highly aggressive malignancy with poor prognoses and frequent resistance to conventional chemotherapy. Approximately 40% of melanoma cases carry the BRAF mutation, for which vemurafenib, a selective BRAF inhibitor, is approved. Despite initial clinical benefits, vemurafenib often leads to drug resistance and relapse, highlighting the need for improved therapeutic strategies.

Astacins, a family of zinc metalloproteinases, are involved in invasion and tissue migration processes in a variety of parasites. An astacin-like proteinases have been detected in the excretory-secretory products (ESPs) of Trichinella spiralis (T. spiralis), zinc metalloproteinase Nas-14 (TsNas14), but its function in T.

Background: This study reveals elevated levels of MMP-1, MMP-2, and MMP-3 in endometriotic lesions, potentially reflecting a senescenceassociated secretory phenotype. No corresponding increase was observed in peritoneal fluid, possibly due to technical or sample-related limitations. These findings support the potential of MMPs as biomarkers and therapeutic targets in endometriosis.

Background/aim: Matrix metalloproteinases (MMPs) are important for extracellular matrix (ECM) degradation with MMP inhibition possibly leading to new antitumor therapies. To examine this possibility, UTX-121, in which the celecoxib sulfonamide was replaced with a methyl ester, was designed to examine the influence of UTX-121 derivatives on MMPs.

Materials And Methods: HT-1080 cells were incubated with UTX-121 derivatives over 48 h, and antitumor activities (IC) were examined using WST-8 assay.
