Protein translation regulation is critical for cellular responses and development, yet how elongation stage disruptions shape these processes remains incompletely understood. Here, we identify a single amino acid substitution (P55Q) in the ribosomal protein RPL-36A of Caenorhabditis elegans that confers complete resistance to the elongation inhibitor cycloheximide (CHX). Heterozygous animals carrying both wild-type RPL-36A and RPL-36A(P55Q) develop normally but show intermediate CHX resistance, indicating a partial dominant effect.

The study of plant biology has traditionally focused on investigations conducted at the tissue, organ, or whole plant level. However, single-cell transcriptomics has recently emerged as an important tool for plant biology, enabling researchers to uncover the expression profiles of individual cell types within a tissue. The application of this tool has revealed new insights into cell-to-cell gene expression heterogeneity and has opened new avenues for research in plant biology.

Motivation: A genome-wide variant effect calibration method was recently developed under the guidelines of the American College of Medical Genetics and Genomics and the Association for Molecular Pathology (ACMG/AMP), following ClinGen recommendations for variant classification. While genome-wide approaches offer clinical utility, emerging evidence highlights the need for gene- and context-specific calibration to improve accuracy. Building on previous work, we have developed an algorithm tailored to converting functional scores from both multiplexed assays of variant effects (MAVEs) and computational variant effect predictors (VEPs) into ACMG/AMP evidence strengths.

Motivation: RNA velocity has become a powerful tool for uncovering transcriptional dynamics in snapshot single-cell data. However, current RNA velocity approaches often assume constant transcriptional rates and treat genes independently with gene-specific times, which may introduce biases and deviate from biological realities. The poor prognosis constitutes a significant difficulty for spinal cord injury (SCI) individuals. Although mesenchymal stem cells (MSCs) hold promises as advanced therapy medicinal products (ATMPs) for SCI patients, challenges such as Good Manufacturing Practice-compliant manufacturing, cellular senescence, and limited therapeutic efficacy continue to hinder their clinical translation. Recent advances have identified botanical nanovesicles (BNs) as potent bioactive mediators capable of "priming" MSCs to self-rejuvenate, augment paracrine effect, and establish inflammatory tolerance.
