Article Abstract

Biosafety Level 4 facilities have been established in our country to promote research and development and national capacity of diagnostic test systems for infectious diseases, including highly pathogenic viruses. In particular, the operation of a suit-type laboratory requiring positive pressure protective suits is a first for our country, and it is necessary to establish unique safety management technology. With the support of the Research Program on Emerging and Re-emerging Infectious Diseases of the Japan Agency for Medical Research and Development (AMED), we conducted research entitled "Study of Practical Biorisk Management in the Microbiological Containment Laboratories" from 2021 to 2023, with the aim of improving biosafety management technology in Japan. We have summarized our research project and its results obtained for the future direction of biorisk management research in Japan.

Introduction: When a life science project is identified as potential dual use research of concern (DURC), United States government policy and biorisk management professionals recommend conducting a risk assessment of the project and using its results to choose strategies to manage any associated risks. However, there is little empirical research on how real-world projects score on DURC assessments, the extent to which reviewers agree or disagree about risks for a given project, or how risk judgments map to recommended risk management strategies. By studying the process of DURC risk assessment, it may be possible to develop methods that are more consistent, accurate, and cost-effective.

Application of a comprehensive approach to pathogen screening in a stowaway rat on an airplane.

In April 2017, a rat was observed on an airplane during a flight from Miami (USA) to Berlin (Germany). After landing in Berlin, significant efforts were made to trap the rat and disinfect the airplane. As rats are known reservoir hosts for a variety of zoonotic pathogens, this event necessitated the establishment of a standard workflow for the detection of rodent-borne pathogens.

Carbon monoxide (CO) is generally recognized as a toxic gas; however, it has recently been identified as an endogenous gasotransmitter with significant cytoprotective properties. CO modulates key molecular pathways, including anti-inflammatory, anti-apoptotic, antioxidant, and vasodilatory signaling pathways, by targeting heme- and non-heme-containing proteins. These proteins include soluble guanylate cyclase, cytochrome P450 enzymes, MAPKs, and NLRP3.

SUMMARYLaboratory-acquired infections (LAIs), particularly those from high-risk viruses, pose significant threats to exposed individuals and to the general public. In this review, we evaluate the existing evidence for viral LAI prevention, including available vaccinations, post-exposure prophylaxis (PEP), and follow-up procedures following occupational exposure to Risk Group 3 and 4 viral infectious agents within clinical testing laboratories. This review provides guidance on the therapeutic options and follow-up, all essential for preparedness planning and timely management in the event of exposure.

Alphaherpesviruses, such as Herpes Simplex Virus 1 (HSV-1) and Pseudorabies Virus (PrV), exhibit pronounced neurotropism. HSV-1 can cause Herpes simplex encephalitis (HSE), primarily affecting the mesiotemporal lobe. CD1 mice inoculated with a PrV mutant (PrV-ΔUL21/US3Δkin) showed striking analogies to HSE, including comparable virus distribution and inflammatory patterns, providing a suitable model to analyze the specificity of the invasion route of alphaherpesviruses.
